高通量测序技术对胚胎停育患者阴道真菌的研究

摘要 目的通过高通量测序技术探究妊娠早期胚胎停育患者阴道真菌分布,比较其与正常早期妊娠者阴道真菌之间的差异,揭示胚胎停育与阴道真菌变化的相关性。方法选取2018年5月至2018年11月在兰州大学第二医院产科就诊的20例患者为研究对象,其中胚胎停育10例(实验组),正常早期妊娠10例(对照组)。分别取两组对象阴道分泌物样本行ITS rDNA高通量测序,测序数据进行OTU聚类分析,Alpha多样性和Beta多样性分析,组间群落差异性分析。结果实验组女性阴道真菌群落中未发现特征菌的存在,但菌群丰度显著高于对照组(P<0.05)。Kazachstania、Rhizopus和Rhodotorula为两组间差异最显著的3个菌属(P<0.05),且它们在实验组中相对丰度均高于对照组。结论阴道真菌丰度的显著变化可能与胚胎停育相关,尤其是Rhizopus丰度显著升高应当引起注意。 Objective To explore the distribution of vaginal mycobiome in patients with missed abortion in early pregnancy,compare the differences between them and the vaginal flora of normal early pregnancy,and reveal the correlation between missed abortion and changes of vaginal fungi.Methods A total of 20 obstetric patients in our hospital from May 2018 to November 2018 were enrolled,including 10 patients in the missed abortion group(Observation group)and 10 patients in the normal early pregnancy group(control group).Vaginal secretion samples were taken for ITS rDNA high-throughput sequencing.The sequencing data were analyzed by using OTU cluster analysis,Alpha diversity and Beta diversity analysis and inter-group community difference analysis to explore the structures of and differences between the two groups of flora.Results No characteristic bacteria were found in the fungal colonies of the observation group,but the abundance of fungi was significantly higher than that of the normal control group(P<0.05).Kazachstania,Rhizopus and Rhodotorula were the three fungal genera with the most significant differences between the two groups(P<0.05),and the relative abundances of the three fungal genera in the observation group were higher than those in the control group.Conclusion Significant changes in the abundance of vaginal fungal flora may be related to missed abortion,especially the significant increase in Rhizopus abundance which should be given special attention.
