摘要
目的探讨头穴丛刺对血管性痴呆大鼠脑组织内质网应激相关因子表达的影响。方法将SD大鼠随机分为空白组、假手术组、模型组和头穴丛刺组,各12只。采用双血管阻断制备大鼠VaD模型。头穴丛刺组给予头穴丛刺针刺治疗,每日1次,连续治疗28d。HE染色光镜观察海马组织形态学的变化;Western blot法检测p-PERK、p-eIF2α蛋白表达。结果模型组大鼠海马神经细胞排列紊乱,可见大量神经元凋亡小体和炎性细胞,毛细血管肿胀;与模型组比较,头穴丛刺组大鼠海马组织神经元细胞核固缩数目减少,胶质细胞、炎细胞和凋亡小体数目减少。与假手术组比较,模型组p-PERK、p-eIF2α蛋白表达量显著升高(P<0.01);与模型组比较,头穴丛刺组p-PERK、p-eIF2α蛋白表达量明显降低(P<0.01)。结论头穴丛刺能够减轻VaD大鼠海马组织炎性反应,改善病变程度,可能与下调p-PERK、p-eIF2α蛋白表达有关。
Objective To investigate the effect of acupuncture on the expression of stress related factors of endoplasmic reticulum in brain tissue of vascular dementia rats.Methods SD rats were randomly divided into blank group,sham group,model group and cluster acupuncture group with 12 rats in each group.The VaD model of rats was prepared by double vessel occlusion.In the group of acupoint cluster acupuncture,acupoint cluster acupuncture was given once a day for 28 consecutive days.The morphological changes of hippocampus were observed by HE staining light microscope.Western blot analysis of p-PERK,p-eIF2α protein expression.Results In the model group,hippocampal nerve cells were disordered,a large number of neuronal apoptotic bodies and inflammatory cells were observed,and the capillaries were swollen.Compared with the model group,the number of neurons in hippocampus decreased,and the number of glial cells,inflammatory cells and apoptotic bodies decreased.The expression levels of p-PERK and p-eIF2α protein in the model group were significantly higher than those in the sham group (P<0.01).Compared with the model group,p-PERK and p-eIF2α protein expressions were significantly decreased in the group (P<0.01).Conclusion Head acupuncture point cluster acupuncture can alleviate the inflammatory response in the hippocampus of VaD rats and improve the degree of lesion,which may be related to the down-regulation of p-PERK and p-eIF2α protein expressions.
