莪术醇诱导人卵巢癌SKOV3细胞凋亡及对PI3K/Akt信号通路相关蛋白的影响 被引量:17

Curcumol Induced Apoptosis of Human Ovarian Cancer SKOV3 Cells and Its Effect on PI3K-Akt Signaling Pathway Related Proteins

摘要 目的观察莪术醇对人卵巢癌SKOV3细胞的凋亡作用及对PI3激酶(PI3K)/丝氨酸/苏氨酸激酶(Akt)信号通路相关蛋白的影响。方法采用MTT法检测不同浓度莪术醇以及临床常用抗肿瘤药物顺铂对人卵巢癌SKOV3细胞的生长抑制作用;流式细胞仪检测细胞凋亡率及对细胞周期的影响;Western blotting法检测莪术醇、顺铂及其两者联合诱导人卵巢癌SKOV3细胞凋亡过程中PI3K/Akt信号通路蛋白(PI3K、p-PI3K、Akt、pAkt)的表达。结果莪术醇对人卵巢癌SKOV3细胞具有生殖抑制作用,其生殖抑制作用呈剂量-时间正效应关系;莪术醇可以促进人卵巢癌SKOV3细胞的凋亡,细胞凋亡率随着莪术醇浓度增大而增强;不同质量浓度莪术醇处理SKOV3细胞48 h后,细胞周期分布发生明显改变,药物的作用将肿瘤细胞的生长阻止在G0/G1和S期;莪术醇、顺铂及其两者联合在诱导人卵巢癌SKOV3细胞凋亡时,下调了PI3K/Akt信号通路中p-PI3K、p-Akt蛋白的表达水平,且莪术醇和顺铂联合应用时具有协同效应,联合组p-PI3K、p-Akt蛋白表达下调最为明显。结论莪术醇可能通过抑制PI3K/Akt信号通路的激活来阻滞人卵巢癌SKOV3细胞周期,诱导细胞凋亡,进而阻止肿瘤细胞的生长或诱导其分化。 Objective:To investigate the effects of Curcumol on apoptosis of human ovarian cancer SKOV3 cells and and its effect on PI3 K-Akt signaling pathway related proteins. Methods:MTT method was used to detect the inhibitory effects of different concentrations of Curcumoland Cisplatin on the growth of human ovarian cancer SKOV3 cells,the flow cytometry(FCM)to determine apoptotic rates of SKOV3 cells and its effect on cell cycle subjected to these treatments,Western blotting to detect the expression of PI3 K/Akt signaling pathway proteins(PI3 K,p-PI3 K,Akt,p-Akt)during the apoptosis of human ovarian cancer SKOV3 cells induced by Curcumol,Cisplatin and their combination. Results:Curcumol could inhibit the reproduction of human ovarian cancer SKOV3 cells in a dose time positive manner. Curcumol could promote the apoptosis of human ovarian cancer SKOV3 cells,and the apoptosis rate increases with its concentration. After the SKOV3 cells were treated with Curcumol at different concentrations for 48 h,the cell cycle distribution changed significantly. The growth of SKOV3 cells was inhibited in G0/G1 and S phase by the effect of Curcumol. Curcumol,Cisplatin and their combination could down regulate the expression of p-PI3 K and p-Akt in PI3 K/Akt signaling pathway when they induce apoptosis of human ovarian cancer SKOV3 cells,and the combination of Curcumol and Cisplatin had synergistic effect,and the expression of p-PI3 K and p-Akt in combination group was the most obvious. Conclusion:Curcumol may inhibit the activation of PI3 K/Akt signaling pathway to block the cell cycle of human ovarian cancer SKOV3 cells,induce apoptosis,and then prevent the growth of tumor cells or induce their differentiation.
