过表达TLR4基因绵羊对口蹄疫疫苗(O型)免疫效应研究 被引量:1

摘要 利用过表达TLR4转基因绵羊模型,注射口蹄疫疫苗(O型),建立口蹄疫灭活疫苗(O型)免疫模型,通过TLR4表达水平、血常规、T淋巴细胞亚群分群、口蹄疫特异性抗体水平,阐明过表达TLR4基因绵羊(n=7)和对照绵羊(n=7)血液中免疫细胞组成和抗体水平动态变化趋势。研究发现,免疫后21 d内,TLR4基因表达水平先降后升;非转基因组白细胞数量逐渐减少,转基因组白细胞数量呈现波动变化,但两组间差异不显著(P>0.05);免疫后淋巴细胞占比略降,单核细胞、中性粒细胞占比在转基因组与非转基因组间差异不显著(P<0.05);免疫后第14天,转基因组CD4^+/CD8^+细胞比值显著高于非转基因组(P<0.05);免疫后49 d内转基因组抗体水平略高于非转基因组,但未达差异显著(P>0.05);研究结果将为解析TLR4基因在口蹄疫免疫应答过程中作用提供理论依据。 A transgenic sheep model that overexpressed TLR4 was injected foot-and-mouth disease vaccine(type O)to establish an inactivated foot-and-mouth disease vaccine(type O)immune model.The expression level of TLR4,blood routine,T lymphocyte subgroup classification and foot-andmouth disease specificity antibody level were tested to clarify the dynamic change trend of immune cell composition and antibody level in the blood of sheep overexpressing TLR4 gene(n=7)and control sheep(n=7).The results showed that within 21 days after immunization,the expression level of TLR4 gene decreased firstly,and then continued to increase;after immunization,the number of white blood cells in the control group gradually decreased,and in the transgenic group showed fluctuations,but the difference between the two groups was not significant(P>0.05);the proportion of lymphocytes decreased slightly after immunization,the proportion of monocytes and neutrophils increased gradually,the difference between the transgenic and the control groups was not significant(P>0.05)after immunization.On the 14th day,the proportion of CD4^+/CD8^+cells in the transgenic group was significantly higher than that in the control group(P<0.05);at each detection time point within 49 days after immunization,the antibody level of the transgenic group was always slightly higher than that of the non-transgenic group;however,there was no significant difference(P>0.05).The results of this study would provide a scientific basis for the analysis of the role of TLR4 gene in the immune response of FMD.
