福建省泉州地区汉族人群TCF7L2 rs11196205基因多态性与T2DM的相关性研究 被引量:2

Correlation between TCF7L2 rs11196205 Gene Polymorphism and Onset Risk of T2DM in Han Population of Quanzhou,Fujian Province

摘要 目的探究转录因子7类似物2(transcription factor 7 like 2,TCF7L2)基因rs11196205多态性与2型糖尿病(type 2 diabetes,T2DM)发病风险的相关性。方法选取2017~2018年福建省泉州地区临床患者和健康体检人群300例作为研究对象,其中健康对照组100例,T2DM组200例。其诊断标准参照中国人群糖尿病诊断标准。所有样本采用提取全血DNA后进行PCR扩增,对TCF7L2 rs11196205位点基因型进行测序。高效液相色谱法检测糖化血红蛋白(hemoglobin Alc,HbAlc)。全自动生化分析仪测定血清中TC,TG,HDL-C,LDL-C,ApoA和ApoB。采用SPSS18.0软件进行数据整理和统计学分析。结果T2DM组GC基因型频率低于正常对照组(χ2=5.053,P<0.05),T2DM组携带C等位基因的频率低于对照组(χ2=4.923,P<0.05),差异有统计学意义。在不同基因型间对照组GC基因型的HbAlc水平低于GG基因型,差异有统计学意义(P=0.031),其余指标在对照组与T2DM组中GC与GG基因型的分布差异均无统计学意义(P>0.05)。二分类Logistic回归结果显示,BMI,HbAlc和GLU是T2DM的独立危险因素,而rs11196205基因型非T2DM的独立危险因素。结论TCF7L2 rs11196205基因多态性与T2DM发病风险相关,C等位基因可能降低T2DM的患病风险,原因可能与C等位基因的血糖水平较低有关。 Objective To explore the relationship between TCF7L2 rs11196205 polymorphism and the risk of type 2 diabetes mellitus(T2DM).Methods From 2017 to 2018,300 cases of clinical patients and healthy people in Quanzhou area of Fujian Province were selected as the research objects,including 100 cases in the healthy control group and 200 cases in the T2DM group.The diagnostic standard refered to the diagnostic criteria for diabetes in the Chinese population.Peripheral blood DNA was extracted and the TCF7L2 rs11196205 gene was amplified by PCR and sequenced SNP.HbAlc was detected by high performance liquid chromatography.Serum levels of TC,TG,HDL-C,LDL-C,ApoA and ApoB were determined by automatic biochemical analyzer.HbAlc level of GC genotype in control group was lower Statistical analysis was performed using SPSS18.0 software.Results The GC genotype frequency of T2DM group was significantly lower than that of normal control group(χ2=5.053,P<0.05),and the frequency of C allele in T2DM group was lower than that in normal control group(χ2=4.923,P<0.05),the difference was statistically significant.The blood than that of GG genotype,the difference was statistically significant(P=0.031),and the remaining indicators were not statistically significant.The results of binary logistic regression data demonstrated that BMI,HbAlc,and GLU were acting as independent risk factors for T2DM,while rs11196205 genotype was not.Conclusion The TCF7L2 rs11196205 gene polymorphism was associated with the risk of T2DM and the C allele may reduce the risk of T2DM.
