摘要
目的探讨新诊断TIDM患者外周血单核细胞(PBMCs)中miR-146a的表达水平与辅助性T细胞17(Th17)、IL-17、IL-6的相关性。方法选取2013年1月至2016年1月于南京医科大学康达学院附属医院内分泌科住院治疗的新诊断TIDM患者68例(TIDM组),另选取同期健康体检者30名为正常对照(NC)组。应用实时荧光定量聚合酶反应测定PBMCs中miR-146a的表达,流式细胞术检测PBMCs中Th17细胞比例,ELISA法测定血浆IL-17、IL-6、TNF-α水平,分析Th17细胞比例与miR-146a、炎性因子等的相关性。结果与NC组比较,TIDM组PBMCs中miR-146a表达水平下降[(2.04±0.72)vs(-1.39±0.499),P<0.05],Th17细胞比例、IL-17、IL-6及TNF-α升高[(1.88±0.09)%vs(4.23±0.86)%,(2.51±0.34)vs(4.77±0.65)ng/L,(3.87±1.78)vs(5.98±2.43)ng/L,(8.28±2.76)vs(13.93±3.53)pg/ml,P<0.05]。Pearson相关分析显示,新诊断T1DM患者Th17细胞比例与IL-17、IL6、TNF-α呈正相关,与miR-146a表达呈负相关(P<0.05)。多元逐步回归分析结果显示,IL-6、miR-146a是新诊断T1DM患者Th17细胞比例的影响因素(P<0.05)。结论T1DM患者外周血miR-146α与Th17细胞比例存在相关性。
ObjectiveTo explore the association between mi R-146 and Th17 cell subset from peripheral blood mononuclear cells(PBMCs)in patients with type 1 diabetes(T1 DM).MethodsA total of 68 hospitalized patients with T1 DM(T1 DM group)and 30 healthy volunteers(NC group)were enrolled in this study from The First Hospital of Lianyungang during January 2013 to January 2016.The expression levels of mi R-146 a in PBMCs were measured by quantitative real-time quantitative polymerase chain reaction(q RT-PCT),and the frequency of Th17 cells in PBMCs was tested by flow sytometry.IL-17、IL-6 were detected by enzyme-linked immunosorbent assay(ELISA).The correlation between mi R-146 a expression and Th17 and inflammatory factors were analyzed.ResultsThe expression levels of mi R-146 a were significantly decreased[(2.04±0.72)vs(-1.39±0.499),P<0.05],the proportion of Th17 cells were significant higher,IL-17,IL-6 and TNF-a were all significantly increased[(1.88±0.09)%vs(4.23±0.86)%,(2.51±0.34)vs(4.77±0.65)ng/L,(3.87±1.78)vs(5.98±2.43)ng/L,(8.28±2.76)vs(13.93±3.53)pg/ml,P<0.05]in T1 DM group than in NC group.Pearson correlation analysis showed that there was a negative association between Th17 cell and mi R-146 a(P<0.05),while there was positive association between Th17 cell and IL-17,IL-6,and TNF-a(P<0.05).Multiple regression analysis indicated that mi R-146 a and IL-6 were independently correlated with Th17(P<0.05).ConclusionThis studysuggested that there might be a correlation between mi R-146 a and Th17 cell subsets in PBMCs in patients with T1 DM.
