摘要
目的探讨GANT61对小鼠关节炎的影响。方法 Balb/c小鼠随机分为3组:正常对照组、胶原诱导的关节炎(CIA)组和GANT61组。CIA组和GANT61组运用牛Ⅱ型胶原诱发小鼠关节炎模型,GANT61组于二次免疫后第0、3、6、9、12、15、18、21天尾静脉注射GANT61。同时观察四肢关节及脚掌情况,并进行临床评分。于初免后第39天,处死小鼠,收集外周血及血清,检测中性粒细胞及细胞因子变化,收集一侧关节,病理学检查,收集另一侧关节滑膜组织,培养滑膜成纤维细胞。流式细胞术及CCK-8法检测细胞增殖情况;流式细胞术检测细胞凋亡情况。结果 GANT61能够减轻关节肿胀程度,降低关节临床评分,同时关节炎症细胞浸润程度降低。中性粒细胞数量降低,细胞因子γ干扰素(IFN-γ)、白细胞介素-6(IL-6)及肿瘤坏死因子α(TNF-α)含量也低于CIA组。滑膜细胞增殖程度降低,凋亡数量增加。结论 GANT61能够通过抑制滑膜细胞增殖,减轻Balb/c小鼠关节炎病理进程。
Objective To explore the effect of GANT 61 on collagen induced arthritis(CIA)mice.Methods BALB/C mice were randomly divided into three groups[Control group,collagen induced arthritis(CIA)group,GANT61 group].CIA group and GANT61 group were induced arthritis with bovine typeⅡcollagen.GANT61 group was injected with GANT61 on 0,3,6,9,12,15,18,21 d after second immunization.Meanwhile the con-dition of joints was observed and the clinical score was made.On 39 th day after primary immunization,mice were executed to collect peripheral blood and serum for detecting the change of neutrophils and cytokines.Joints and synovial tissues were collected for pathological examination and synovioblast culture.Cell proliferation was detected by flow cytometry and CCK-8 assay.Apoptosis was detected by flow cytometry.Results Data showed that GANT61 could reduce joint swellingand clinical score of arthritis,the infiltration of inflammatory cells can also be reduced.Neutrophils and inflammation cytokines of interferon-gamma(IFN-γ),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α).GANT61 could remarkably inhibit cell proliferation and induce cell apoptosis.Conclusion GANT61 can alleviate the pathological progress of arthritis in Balb/c mice by inhibiting the proliferation of synovial fibro-blasts.
