电针预处理对心肌缺血大鼠心肌自噬相关蛋白LC3Ⅰ、LC3Ⅱ、Beclin 1、HIF-1α表达影响 被引量:5

Effect of Electroacupuncture Pretreatment on Expession of Autophagy Related Proteins LC3Ⅰ,LC3Ⅱ,Beclin 1,HIF-1αin Rats with Myocardial Ischemia

摘要 目的观察电针内关预处理对心肌缺血(AMI)大鼠心肌自噬相关蛋白的影响。方法大鼠随机分成伪手术组、模型组、电针预处理组;构建AMI大鼠模型,造模前电针刺激14 d,刺激电流1 mA,频率为2 Hz,30 min/d。运用PowerLab 16导生理记录仪记录分析各组大鼠心电图,Western Blot检测心肌组织微管相关蛋白轻链3Ⅰ(LC3Ⅰ)、微管相关蛋白轻链3Ⅱ(LC3Ⅱ)、Beclin 1基因(Beclin 1)、缺氧诱导因子-1α(HIF-1α)水平,PCR检测LC3ⅠmRNA、LC3ⅡmRNA、Beclin 1 mRNA、HIF-1ɑmRNA。结果与伪手术组比较,模型组LC3Ⅱ、Beclin 1蛋白及mRNA表达水平升高,LC3Ⅱ/LC3Ⅰ比值上升(P<0.01),LC3Ⅰ、HIF-1α蛋白及mRNA表达水平显著下降(P<0.01);与模型组相比,电针组LC3Ⅱ、Beclin 1蛋白及mRNA表达水平明显下降,LC3Ⅱ/LC3Ⅰ比值降低(P<0.01),LC3Ⅰ、HIF-1α蛋白及mRNA水平升高(P<0.01)。结论针刺预处理对AMI大鼠心肌保护作用可能与下调LC3Ⅱ、Beclin 1及LC3Ⅱ/LC3Ⅰ比值,上调LC3Ⅰ、HIF-1α表达有关。 Objective To observe the effect of electroacupuncture(EA)pretreatment at Neiguan on the expression of LC3Ⅰ,LC3Ⅱ,Beclin 1,HIF-1αin myocardial cells of rats with acute myocardial ischemia(AMI).Methods Rats were randomly divided into sham-operated group,model group and EA pretreatment group.The AMI model was established.Before modeling,EA pretreatment was continuously stimulated the PC6 for 30 min,once daily for 14 days,and the stimulating current was 1 mA,the frequency was 2 Hz.The electrocardiogram of rats in each group was recorded and analyzed by powerlab 16-channel physiological recorder.The expression of autophagy protein LC3Ⅰ,LC3Ⅱ,Beclin 1,HIF-1αwas detected by Western Blot.The expression levels of LC3ⅠmRNA,LC3ⅡmRNA,Beclin 1 mRNA,HIF-1αmRNA were detected by PCR.Results Compared with the sham groups,the expression levels of autophagy protein LC3Ⅱ,Beclin 1,LC3ⅡmRNA,Beclin 1 mRNA and the ratio of LC3Ⅱ/LC3Ⅰwere significantly increased(P<0.01),the expression levels of LC3Ⅰ,HIF-1α,LC3ⅠmRNA,HIF-1αmRNA were significantly decreased(P<0.01);Compared with model group,the expression levels of autophagy protein LC3Ⅱ,Beclin 1,LC3ⅡmRNA,Beclin 1 mRNA and the ratio of LC3Ⅱ/LC3Ⅰwere significantly decreased,the expression levels of LC3Ⅰ,HIF-1α,LC3ⅠmRNA,HIF-1αmRNA were significantly increased(P<0.01).Conclusion EA pretreatment of Neiguan has a protective effect on myocardial ischemia,which is probably related to the effect in regulating expression of LC3Ⅱ,Beclin 1,the ratio of LC3Ⅱ/LC3Ⅰand LC3Ⅰ,HIF-1α.
