摘要
目的探究运动疗法对链脲佐菌素(STZ)诱导的糖尿病大鼠病理损伤及骨骼肌叉头框转录因子O亚族1(FOXO1)、葡萄糖转运子4(GLUT4)表达的影响。方法选取75只雄性Wistar大鼠,随机选取60只制成糖尿病模型,并将其分为模型组、低强度运动组、中等强度运动组、高强度运动组,剩余15只作为健康对照组;使用HE染色观察其腓肠肌;使用蛋白质印记检测含半胱氨酸的天冬氨酸蛋白水解酶-3(Caspase-3)、含半胱氨酸的天冬氨酸蛋白水解酶-9(Caspase-9)、、FOXO1及GLUT4蛋白表达情况;使用全自动生化分析仪检测血糖、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白-胆固醇(HDL)、低密度脂蛋白-胆固醇(LDL)水平;使用酶联免疫吸附法检测白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)水平;使用生物化学方法测定主动脉血管组织匀浆中组织超氧化物歧化酶(SOD)、丙二醛(MDA)、乳酸脱氢酶(LDH)的活性水平;使用RT-PCR检测FOXO1、GLUT4 mRNA表达水平。结果与健康对照组相比,模型组大鼠Caspase-3、Caspase-9、FOXO1蛋白表达、TC、TG、LDL、L-6、TNF-α、FOXO1 mRNA表达水平、LDH、MDA活性升高(P<0.05),GLUT4蛋白表达、HDL、IL-10、SOD及GLUT4 mRNA表达水平降低(P<0.05);与模型组相比,运动各组大鼠Caspase-3、Caspase-9、FOXO1蛋白表达、TC、TG、LDL、IL-6、TNF-α、FOXO1mRNA表达水平、乳酸脱氢酶(LDH)、MDA活性降低(P<0.05)且中等强度运动组上述指标最低,GLUT4蛋白表达、HDL、IL-10、SOD及GLUT4 mRNA表达水平升高(P<0.05)且中等强度运动组上述指标最高。结论运动疗法可以通过升高骨骼肌GLUT4蛋白及mRNA表达水平、降低骨骼肌FOXO1蛋白及mRNA表达水平缓解糖尿病大鼠病理损伤,且中等运动强度效果最佳。
Objective To explore the effects of exercise therapy on pathological injury and expression of skeletal muscle forkhead box transcription factor O1(FOXO1) and glucose transporter 4(GLUT4) in diabetes rats induced by streptozotocin(STZ). Methods A total of 75 male Wistar rats were enrolled and 60 cases of them were randomly selected and made into diabetes model, and they were divided into model group, low-intensity, middle-intensity and high-intensity exercise groups.The remaining 15 rats were enrolled as healthy control group.HE-staining was applied to observe gastrocnemius muscle.The expression of cysteinyl aspartate specific proteinase-3(Caspase-3),cysteinyl aspartate specific proteinase-9(Caspase-9),FOXO1 and GLUT4 proteins was detected by Western blot.The levels of blood glucose, total cholesterol(TC), triglyceride(TG), high density lipoprotein-cholesterol(HDL) and low density lipoprotein-cholesterol(LDL) were detected by automatic biochemical analyzer. The enzyme-linked immunosorbent assay was applied to detect levels of interleukin-6(IL-6), tumor necrosis factor-α ( TNF-α) and interleukin-10( IL-10). The biochemical methods were applied to determine activity levels of superoxide dismutase( SOD),malondialdehyde( MDA) and lactate dehydrogenase( LDH) in tissue homogenate of aortic vessel. The expression levels of FOXO1 and GLUT4 mRNA were detected by RT-PCR. Results Compared with healthy control group,expression of Caspase-3,Caspase-9 and FOXO1 protein,expression levels of TC,TG,LDL,IL-6,TNF-α and FOXO1 mRNA,activity of LDH and MDA were increased in model group( P < 0. 05),while expression of GLUT4 protein,expression levels of HDL,IL-10,SOD and GLUT4 mRNA were decreased( P< 0. 05). Compared with model group,expression of Caspase-3,Caspase-9 and FOXO1 protein,expression levels of TC,TG,LDL,IL-6,TNF-α and FOXO1 mRNA,activity of LDH and MDA were decreased in all exercise groups( P < 0. 05). And the above indexes were the lowest in medium-intensity exercise group,while GLUT4 protein expression,expression levels of HDL,IL-10,SOD and GLUT4 mRNA were increased( P < 0. 05). And the above indexes were the highest in medium-intensity exercise group. Conclusion Exercise therapy can alleviate pathological injury of diabetes rats by increasing expression levels of skeletal muscle GLUT4 protein and mRNA,and decreasing expression levels of skeletal muscle FOXO1 protein and mRNA. The effect of middle-intensity exercise is the best.
