摘要
目的观察和比较刺山柑果风湿止痛贴和温和灸对佐剂性关节炎大鼠滑膜组织免疫蛋白HSP70和PPARγ表达的影响,探讨其可能的免疫调节作用机制。方法50只大鼠随机分为正常组、模型组、阳性药组、刺山柑组和艾灸组。采用足跖皮下注射弗氏完全佐剂建立佐剂性关节炎大鼠模型。阳性药组和刺山柑组分别用伤湿止痛膏和刺山柑果风湿止痛贴外敷,艾灸组进行温和悬灸肾俞、足三里。观察大鼠AI指数和足爪容积、血清TNF-α和IL-1β水平、滑膜组织HSP70和PPARγ蛋白表达。结果造模后各组大鼠关节肿胀明显,TNF-α、IL-1β水平和滑膜组织HSP 70、PPARγ蛋白表达均显著升高(P<0.05或0.01)。治疗3个疗程后,刺山柑组和艾灸组大鼠血清中TNF-α、IL-1β水平均明显降低(P<0.05);艾灸组大鼠HSP70表达明显升高(P<0.05)并明显高于刺山柑组(P<0.01);刺山柑组PPARγ蛋白表达明显升高(P<0.01)并高于艾灸组(P<0.05)。结论刺山柑和艾灸疗法均可下调抗炎因子TNF-α、IL-1β水平,其作用机制可能是刺山柑上调PPARγ的表达、艾灸上调HSP70的表达而发挥抗炎消肿止痛作用。
Objective To observe and compare the effects of Capparis Spinosa Fruits Pain Gel Paste and mild moxibustion of HSP70 and PPARγin synovial tissue of adjuvant arthritis rats,and to explore the mechanism of their effects on immune regulation.Methods A total of 50 rats were randomly divided into normal group,model group,positive drug group,Capparis Spinosa group and moxibustion group.The model of rats was established by subcutaneous injection of Freund's complete adjuvant into the sole of the foot.The positive drug group and Capparis Spinosa group were applied with Shangshi Zhitong Gao and Capparis Spinosa Fruits Pain Gel Paste respectively.The moxibustion group was treated with mild suspension moxibustion with Shenshu(BL 23)and Zusanli(ST 36).AI index,foot volume,serum levels of TNF-αand IL-1β,expression of HSP70 and PPARγin synovial tissue were observed.Results The swelling of the joints was obvious,the levels of TNF-α,IL-1βand the expressions of HSP70 and PPARγin synovial tissue were significantly increased(P<0.05 or 0.01).After treatment,the levels of TNF-αand IL-1βin serum of rats of the Capparis Spinosa group and moxibustion group decreased significantly(P<0.05);the expression of HSP70 in the moxibustion group increased significantly(P<0.05)and was significantly higher than that in the Capparis Spinosa group(P<0.01);the expression of PPARγin the Capparis Spinosa group increased significantly(P<0.01)and was significantly higher than that in the moxibustion group(P<0.05).Conclusion Both Capparis Spinosa and moxibustion therapy can down-regulate the levels of anti-inflammatory factors TNF-αand IL-1β.The possible mechanism is that Capparis Spinosa can up-regulate the expression of PPARγand moxibustion can up-regulate the expression of HSP70 and plays an anti-inflammatory,anti-swelling and analgesic role.
