miRNA155在阿霉素诱导乳腺癌上皮间质转化过程中的表达及其作用

摘要 目的:观察阿霉素对乳腺癌MCF-7细胞上皮间质转化(EMT)及miRNA表达的影响,并观察瞬转miRNA155对MCF-7细胞凋亡及侵袭的影响。方法:采用10 ng/mL阿霉素与乳腺癌MCF-7细胞体外共培养1周、2周、3周,观察细胞形态变化,western blot、免疫荧光检测各组细胞E-cadherin表达,RT-PCR检测miRNA155、miRNA200c的表达情况。进一步通过流式细胞术、Transwell实验检测miRNA155对MCF-7细胞凋亡及侵袭能力的影响。结果:与对照组相比,各阿霉素处理组细胞E-cadherin表达均低于对照组,随处理时间增加,E-cadherin表达逐渐降低(均P<0.05)。阿霉素处理后miRNA155表达水平显著增高(P<0.05),miRNA200c表达无明显变化(P>0.05)。miRNA155过表达组凋亡峰和细胞穿透数小于对照组及空转染组(均P<0.05)。结论:阿霉素可诱导乳腺癌MCF-7细胞发生EMT,miRNA155可能参与介导此过程,上调miRNA155可能增加阿霉素的耐药性,但却抑制乳腺癌MCF-7细胞的侵袭能力。 Objective:To observe the effects of adriamycin on epithelial mesenchymal transition(EMT)and miRNA expression in breast cancer MCF-7 cells.And to view the effect of miRNA155 on apoptosis and invasion of MCF-7 cells.Methods:Breast cancer MCF-7 cells were cultured with adriamycin at a dose of 10 ng/mL for 1 week,2 weeks,and 3 weeks,respectively.The morphological changes of MCF-7 cells under different intervention durations were observed.Western blot and immunofluorescence were used to detect the expression of E-cadherin.The expressions of miRNA155 and miRNA200c were detected by RT-PCR.The effect of miRNA155 on MCF-7 cell apoptosis and invasion ability were further detected by flow cytometry and transwell experiment respectively.Results:Compared with the control group,the E-cadherin expression in adriamycin group was reduced.With the increase of adriamycin treatment time,the E-cadherin expression was gradually decreased(all P<0.05).Moreover,adriamycin induced significant increase of miRNA155(P<0.05),without obvious change in miRNA200c(P>0.05),when compared to the control group.The apoptotic peak and the cell penetration number in the miRNA155 transfection group was decreased when compared to the control group and the empty transfection group(all P<0.05).Conclusion:Adriamycin can induce the EMT in breast cancer MCF-7 cells,and miRNA155 may be involved in this process.Up-regulation of miRNA155 may increase the resistance of adriamycin,but inhibit the invasion ability of breast cancer MCF-7 cells.
