摘要
为了找出新疆某羊场羔羊陆续发病死亡的原因,试验解剖送检的6只羔羊,对组织病料进行细菌分离培养、染色鉴定、16S rRNA PCR鉴定、测序比对分析,并用分离株进行动物致病性试验和药敏试验。结果表明:病原菌在BHI琼脂培养基上生长情况良好,为灰色不透明小菌落和透明扁平菌落,革兰氏染色为阳性球菌。PCR扩增出的条带大小为1 470 bp,将病原菌16S rRNA序列提交至GenBank中,与金黄色葡萄球菌和粪链球菌核苷酸序列的同源性分别为99%和98%。分离得到的金黄色葡萄球菌对小鼠致病性强,10 h内小鼠均死亡,小鼠对粪链球菌不敏感。分离得到的这2株菌对克林霉素都有耐药性;对硫酸庆大霉素、左氧氟沙星、替米考星和乳酸环丙沙星均有敏感性。说明此次羔羊发病是金黄色葡萄球菌与粪链球菌混合感染引起,交替使用4种敏感药物对该地区养殖场发病羊进行治疗与预防可取得一定效果。
In order to find out the reason of the death of lambs in a sheep farm in Xinjiang province, 6 lambs’ tissue samples were dissected and tested. for bacterial isolation and culture, staining and identification, 16 S rRNA PCR identification, sequencing comparison analysis, and animal pathogenicity test and drug sensitivity test were conducted with isolated strains. The results showed that the pathogenic bacteria grew well on BHI agar medium, which were gray opaque small colonies and transparent flat colonies, and gram staining was negative cocci. The PCR amplification band size was 1 470 bp, and the 16 S rRNA sequence of the pathogen was submitted to GenBank, and the nucleotide sequence homology with Staphylococcus aureus and Streptococcus faecalis was 99% and 98%, respectively. Staphylococcus aureus isolates were highly pathogenic to mice, all of them die within 10 hours, while the mice were not sensitive to Streptococcus faecalis. The two strains isolated were all resistant to clindamycin. They were sensitive to gentamicin sulfate, levofloxacin, tilmicosin and ciprofloxacin lactate. This indicated that the lamb’s onset was caused by the mixed infection of Staphylococcus aureus and Streptococcus faecalis. The use of four kinds of sensitive drugs to treat and prevent the sheep on the farms in the area could achieve certain results.
