RAF/MEK/ERK信号通路在胰腺癌发生发展中的作用机制及其临床应用价值 被引量:2

摘要 丝裂原活化蛋白激酶(MAPK)信号通路的激活在多种肿瘤发生发展过程中起着重要作用。在胰腺癌中,受体酪氨酸激酶的异常活化可以激活MAPK信号通路,在MAPK家族中,细胞外信号调节激酶(RAF/MEK/ERK)信号通路是其中最重要的信号通路,活化的RAF/MEK/ERK信号通路能够传递细胞外信号调节肿瘤的增殖、凋亡、侵袭转移和血管生成。因此,RAF/MEK/ERK信号通路被认为是胰腺癌治疗的潜在治疗靶点。该文重点综述了RAF/MEK/ERK信号通路的异常活化在胰腺癌发生发展中的生物学作用,靶向抑制剂的研发及其在临床应用中存在问题和挑战。 The activation of mitogen-activated protein kinase(MAPK)signaling pathway plays an important role in the development of various tumors.In pancreatic cancer,abnormal activation of receptor tyrosine kinases can activate the MAPK signaling pathway,and in the MAPK family,the extracellular signal-regulated kinase(RAF/MEK/ERK)signaling pathway is the most important of these signaling pathways,and the activated RAF/MEK/ERK signaling pathway is able to transmit extracellular signals to regulate tumor proliferation,apoptosis,invasion and metastasis,and angiogenesis.Therefore,the RAF/MEK/ERK signaling pathway is considered as a potential therapeutic target for pancreatic cancer treatment.This review focuses on the biological role of abnormal activation of the RAF/MEK/ERK signaling pathway in the development of pancreatic cancer,the research and development of targeted inhibitors and their existing problems and challenges in clinical application.
