摘要
目的分析T细胞免疫球蛋白和黏蛋白结构域3(Tim-3)/高迁移率族蛋白1(HMGB1)在布氏杆菌感染者中的表达及其与相关细胞因子水平的关系。方法选取2017年10月—2018年10月于新疆维吾尔自治区人民医院感染科住院的布氏杆菌感染者32例为布病组,于同期医院体检正常者30例为健康对照组,分别测定2组血清Tim-3、HMGB1、肿瘤坏死因子α(TNF-α)、白介素1β(IL-1β)、干细胞因子(SCF)水平,同时计数外周血单个核细胞(PBMC)中转化生长因子-β(TGF-β)的斑点形成细胞数(SFC)。结果布病组Tim-3、HMGB1、TNF-α、IL-1β及SCF水平均高于健康对照组[(13.54±2.02)ng/ml vs.(7.43±1.76)ng/ml,(25.84±6.82)ng/ml vs.(13.23±4.75)ng/ml,(74.54±22.66)pg/ml vs.(37.43±10.38)pg/ml,(205.84±84.82)pg/ml vs.(83.23±25.63)pg/ml,(225.25±76.72)pg/ml vs.(103.43±66.67)pg/ml,t=12.662、8.394、8.196、7.595、6.667,P均<0.01]。布病组中采用布氏杆菌重组蛋白抗原(BCSP31)为实验刺激孔TGF-β的SFC明显多于对照组(t/P=28.590/<0.001),而2组中阴性与阳性对照孔TGF-β的SFC比较,差异无统计学意义(P>0.05)。结论布氏杆菌病患者血清Tim-3/HMGB1水平均增加,且在布氏杆菌抗原刺激后形成TGF-β的SFC明显增加。
Objective To analyze the expression of T cell immunoglobulin and mucin domain 3(Tim3)/high mobility group box 1(HMGB1)in Brucella-infected patients and their relationship with the levels of related cytokines.Methods Thirty-two brucella-infected patients who were hospitalized in the Department of Infectious Diseases,People’s Hospital of Xinjiang Uygur Autonomous Region from October 2017 to October 2018 were selected as the brucellosis group,and 30 patients with normal physical examination in the hospital during the same period were selected as the healthy control group,and the two groups of serum were measured.Tim 3,HMGB1,tumor necrosis factorα(TNF-α),interleukin 1β(IL-1β),stem cell factor(SCF)levels,and count the formation of transforming growth factorβ(TGF-β)spots forming cells(SFC)in peripheral blood mononuclear cells(PBMC).Results The levels of Tim-3,HMGB1,TNF-α,IL-1βand SCF in the brucellosis group were higher than those in the healthy control group[(13.54±2.02)ng/ml vs.(7.43±1.76)ng/ml,(25.84±6.82)ng/ml vs.(13.23±4.75)ng/ml,(74.54±22.66)pg/ml vs.(37.43±10.38)pg/ml,(205.84±84.82)pg/ml vs.(83.23±25.63)pg/ml,(225.25±76.72)pg/ml vs.(103.43±66.67)pg/ml,t=12.662,8.394,8.196,7.595,6.667,all P<0.01].Brucella recombinant protein antigen(BCSP31)was used as the experimental stimulation hole TGF-βSFC in the brucellosis group was significantly more than that of the control group(t/P=28.590/<0.001),while the negative and positive control hole TGF-βin the two groups compared with SFC,the difference was not statistically significant(P>0.05).Conclusion The serum Tim3/HMGB1 level of patients with brucellosis increased,and the SFC forming TGF-βincreased significantly after the stimulation of Brucella antigen.
