摘要
目的探讨不同剂量低能量激光(low level laser irradiation,LLLI)对人牙周膜干细胞(human periodontal mesenchymal stem cells,hPDLSCs)细胞增殖和成骨分化的影响,为下一步LLLI在骨组织修复中的应用提供一定参考。方法将处于对数生长期的hPDLSC分为4组,分别接受照射不同剂量LLLI(Nd:YAG;Wavelength:1064 nm)照射,记为0 J/cm^2组、5 J/cm^2组、10 J/cm^2组和20 J/cm^2组。通过MTT法和克隆形成试验检测细胞增殖,RT-PCR检测成骨分化相关基因Runx2、ALP水平,茜素红矿化结节染色实验检测细胞成骨分化。结果MTT实验显示,与0 J/cm^2组相比,5 J/cm^2组和10 J/cm^2组细胞增殖能力显著增高(P<0.05),20 J/cm^2组显著降低(P<0.05);CFU-F实验显示,10 J/cm^2组细胞克隆形成能力增高(P<0.05),20 J/cm^2组显著下降(P<0.05)。茜素红定量实验和PCR结果均表明,与0 J/cm^2组相比,5 J/cm^2和10 J/cm^2组成骨分化能力显著提高(P<0.05),20 J/cm^2组显著下降(P<0.05)。结论5 J/cm^2的LLLI可以提高hPDLSCs的成骨分化能力,10 J/cm^2的LLLI可以提高hPDLSCs的增殖能力和成骨分化能力。
Objective To explore the effects of low-level laser irradiation(LLLI)at different energy intensity on proliferation and osteogenic differentiation of human periodontal mesenchymal stem cells(hPDLSCs),and to provide a basis for its application in bone tissue repair.Methods The hPDLCs in logarithmic growth phase were divided into four groups and subjected to LLLI(Nd:YAG;Wavelength:1064 nm)at different intensity(0,5,10,20 J/cm^2).Cell proliferation was measured using colony forming unit-fibroblast(CFU-F)assay and MTT assay.Osteogenic capacity of cells was determined by Alizarin red staining and the mRNA transcript levels of genes runt-related transcription factor 2(Runx2),ALP.Results Compared with 0 J/cm^2 group,the cell proliferation ability was increased significantly in 5 J/cm^2 group and 10 J/cm^2 group,but was reduced significantly in 20 J/cm^2 group(P<0.05).The cell clone formation ability was increased significantly in 10 J/cm^2 group and decreased significantly in 20 J/cm^2 group.Both the Alizarin red quantitative experiment and PCR results indicated that compared with 0 J/cm^2,the bone differentiation ability was improved significantly in 5 J/cm^2 group and 10 J/cm^2 group(P<0.05),and was decreased significantly in 20 J/cm^2 group(P<0.05).Conclusion The 5 J/cm^2 LLLI can improve the osteogenic differentiation ability of hPDLSCs,and 10 J/cm^2 LLLI can improve the proliferation and osteogenic differentiation of hPDLSCs.
