摘要
目的:应用斑马鱼高通量筛选平台筛选抗炎中药(民族药),并用小鼠急性炎症模型对其抗炎作用进行验证。方法:应用斑马鱼急性炎症疾病模型(Zlyz-EGFP)活体高通量筛选抑制斑马鱼巨噬细胞迁移、聚集的中药(民族药),并以二甲苯致小鼠耳肿胀、鸡蛋清致小鼠足跖肿胀急性炎症模型验证其抗炎作用。结果:通过高通量筛选发现血人参乙醇提取物和飞龙掌血乙醇提取物可显著抑制斑马鱼巨噬细胞向急性损伤部位迁移、聚集(P<0.05,P<0.01),并显著抑制两种小鼠模型的急性炎症(P<0.01)。结论:血人参乙醇提取物和飞龙掌血乙醇提取物可抑制斑马鱼巨噬细胞向急性损伤部位迁移、聚集,具有显著的抗炎作用,为临床应用提供依据。
Objective:To screen anti-inflammatory Chinese materia medica(Ethnic medicine)by zebrafish highthroughput screening platform,and the anti-inflammatory effect was verified by acute inflammation model in mice.Methods:The acute inflammatory disease model of zebrafish(Zlyz-EGFP)was used for high-throughput screening of Chinese materia medica(ethnic medicine)that inhibit the migration and aggregation of macrophages in zebrafish,and the anti-inflammatory effect was verified by an acute inflammation model of xylene-induced ear swelling in mice and egg-white swelling in mice.Results:High-throughput screening results found that the ethanol extracts from Buerger’s Lespedeza Root and Asiatic Toddalia Root can significantly inhibit the migration and aggregation of zebrafish macrophages to the acute injury site(P<0.05,P<0.01)and acute inflammation in two different mouse models(P<0.01).Conclusion:The ethanol extracts from Buerger’s Lespedeza Root and Asiatic Toddalia Root have significant anti-inflammatory effects,our results can provide a basis for clinical application.
