基于SSR标记的201份康定樱桃种质资源DNA指纹图谱构建 被引量:4

Construction of DNA Fingerprinting with SSR Markers for 201 Kangding Cherry(Cerasus tatsienensis) Accessions

摘要 本研究利用筛选出的25对多态性高、条带清晰的SSR引物,以湖北、四川、云南、河南、山西和陕西201份康定樱桃为材料,进行SSR指纹图谱的构建和遗传多样性分析。结果表明:25对引物在供试材料中共检测出194个等位位点,多态性条带比例为100%,平均Nei’s遗传多样性指数(H)为0.755,平均多态性信息含量(PIC)为0.730;采用引物-带型组合法构建了201份康定樱桃种质的指纹图谱;对201份康定樱桃种质做聚类分析,将其分为三组:Ⅰ组为鄂居群(鹤峰,大九湖,神农顶,官门山,天燕);Ⅱ组为川鄂滇居群(星斗山,雅家梗,海螺沟,亚拉,木林子,玉龙雪山,峨眉山,紫石乡,东拉山);Ⅲ组为秦晋豫居群(红河谷,宝天曼,舜王坪)。本研究成功构建了康定樱桃的SSR指纹图谱,并根据其居群遗传距离作出聚类分析,该结果为后期康定樱桃种质资源鉴定和分子育种提供了重要依据。 The SSR fingerprint map and genetic diversity of 201 Cerasus tatsienensis accessions, which were lived in Hubei, Sichuan, Yunnan, Henan, Shanxi and Shanxi Province, were studied using 25 pairs of selected polymorphic SSR primers. The results indicated that all the primers showed polymorphism, 194 polymorphic alleles were revealed, the average Nei’s genetic diversity index(H) was 0.755, and the average polymorphic information content(PIC) was 0.730. A strategy of combining primer pair with distinct alleles for fingerprint construction was developed and applied to the 201 accessions. The data of 201 tested materials were analyzed by unweighted pair-group method with arithmetic means(UPGMA), showed that Group Ⅰ was GMS, SND, DJH, TY and HF populations.GroupⅡ was MLZ, XDS, YLXS, YJG, HLG, DLS, ZSX, EMS and YL populations. GroupⅢ was BTM, SWP and HHG populations. This paper successfully constructed the SSR fingerprint of Cerasus tatsienensis and made cluster analysis based on its population genetic distance. The DNA fingerprinting provides a technical reference for the varieties identification and molecular breeding in Cerasus tatsienensis.
