基于多指标定量和指纹图谱技术综合评价的正交试验优选活血降糖颗粒水提工艺 被引量:5

Optimization of Extraction Process for Huoxue Jiangtang Granules based on Orthogonal Experimental of Multi-index Quantification and Fingerprint Technology Comprehensive Evaluation

摘要 目的:以多指标综合评分法优选活血降糖颗粒的水提工艺。方法:采用正交试验设计法,以加水量、煎煮次数、煎煮时间为影响因素,梓醇含量、毛蕊异黄酮葡萄糖苷含量、总固形物质量、提取指纹图谱共有峰主成分所得综合评分为评价指标,优化活血降糖颗粒的水提工艺。结果:梓醇和毛蕊异黄酮葡萄糖苷分别在20.230~141.610μg/mL(r^1=0.9999)、0.698~8.376μg/mL(r^2=0.9993)内呈良好的线性关系,加样回收率分别为100.42%、100.46%,RSD均小于3.0%;正交样品指纹图谱共标定了19个共有峰,主成分分析提取了4个主成分;优选的最佳水提工艺为:加8倍量水,煎煮2次,每次1 h。结论:以多指标优选活血降糖颗粒的水提工艺,不仅更客观全面,还能兼顾大多数成分种类与含量的变化情况。 Objective:To optimize the water extraction process of Huoxue jiangtang granules by multi-index comprehensive scoring.Methods:Using the orthogonal experimental design method,the water extraction process of Huoxue jiangtang granules was optimized by taking the amount of water added,the number of times of decocting and the decocting time as the influencing factors,catalpol and calycosin-7-glucoside contents,total solid mass and the comprehensive score of common peak principal components in the extracted fingerprint as the evaluation indexes.Results:The linear relationships of catalpol and calycosin-7-glucoside were good in 20.230~141.610μg/mL(r^1=0.9999)and 0.698~8.376μg/mL(r^2=0.9993),respectively.The recoveries were 100.42%and 100.46%,respectively,with RSD less than 3.0%.A total of nineteen common peaks were identified in the fingerprint and four principal components were extracted by principal component analysis.The optimal water extraction process was to add 8 times of water,extracting twice times,1 h each time.Conclusion:The water extraction technology of Huoxue jiangtang granules is optimized by multiple indexes,which is not only more objective and comprehensive,but also can take into account the changes of most components and contents.
