摘要
目的:探究纳布啡对分娩大鼠PKCε/TRPV1信号通路及镇痛效果的影响。方法:将30只孕鼠随机分为对照组、H-7组、纳布啡组,每组10只。制备硬膜外镇痛模型,分娩开始后分别给予生理盐水、H-7稀释液和纳布啡注射液泵注治疗。通过热水甩尾法、脊髓线粒体功能和还原型谷胱甘肽(GSH)水平观察镇痛效果;ELISA检测血清5-HT水平;免疫组织化学(IHC)检测脊髓k受体(KOR)表达;WB检测蛋白激酶C(Protein Kinase C, PKC)、瞬时受体电位香草酸亚型1(transicent receptor potential vanilloid 1, TRPV1)、TRP锚蛋白1(transicent receptor potential ankyrin 1, TRPA1)相对表达水平。结果:分娩镇痛后H-7组、纳布啡组大鼠痛阈值较对照组显著升高,血清5-HT水平降低(均P<0.01);干预后两组大鼠线粒体呼吸功能较对照组增强,线粒体内GSH水平显著升高(P<0.05,P<0.01);与对照组和H-7组比较,纳布啡组KOR表达量最高(均P<0.05);与对照组比较,H-7组、纳布啡组大鼠脊髓内PKCε、TRPV1、TRPA1蛋白表达受到显著抑制(均P<0.001)。结论:纳布啡对分娩大鼠具有有效镇痛效果,其镇痛机制可能与k受体激活、脊髓线粒体呼吸调节及PKC/TRPV1信号通路抑制有关。
Objective:To explore the effects of nalbuphine on PKCε/TRPV1 signal pathway and analgesia in delivery rats. Methods: A total of 30 pregnant rats were randomly divided into control group, H-7 group and nalbuphine group,with 10 rats in each group. The epidural analgesia model was established. After the beginning of delivery, normal saline, H-7 diluent and nalbuphine injection were given respectively. The analgesic effect was observed by hot water tail flick method, spinal cord mitochondrial function and reduced glutathione(GSH) level.Serum 5-HT level was detected by ELISA.Spinal k receptor(KOR) expression was detected by immunohistochemical(IHC).The relative expression levels of protein kinase C(PKC), transicent receptor potential vanilloid 1(TRPV1) and transicent receptor potential ankyrin 1(TRPA1) were detected by WB.Results:After delivery analgesia, the pain threshold was significantly higher and the level of serum 5-HT was lower in H-7 group and nalbuphine group than those in control group(all P<0.01). After intervention, the respiratory function of mitochondria and the level of GSH in mitochondria in both groups were significantly higher than those in control group(P<0.05, P<0.01).Compared with control group and H-7 group, the expression of KOR in nalbuphine group was the highest(all P<0.05). Compared with the control group, the expression of PKCε, TRPV1 and TRPA1 protein in the spinal cord of H-7 group and nalbuphine group was significantly inhibited(all P<0.001).Conclusion: Nalbuphine has an effective analgesic effect on parturient rats, and its analgesic mechanism may be related to the activation of k receptor, respiratory regulation of spinal cord mitochondria and inhibition of PKC/TRPV1 signal pathway.
