摘要
目的检测人参皂苷Rg1对LPS诱导RAW264.7巨噬细胞炎症因子IL-6和促炎因子lncRNA NEAT1表达的影响。方法RAW264.7巨噬细胞分为对照组、LPS组、LPS+不同浓度(5、50μmol/L)人参皂苷Rg1组。分别在6h后收集各组细胞和培养上清,MTT检测细胞存活率,RT-qPCR检测细胞内IL-6 mRNA和NEAT1表达,ELISA检测上清中细胞因子IL-6的含量。结果与对照组相比,LPS诱导细胞巨噬细胞内IL-6 mRNA和NEAT1表达上调,培养上清中IL-6分泌增加,细胞存活率下降(均P<0.05);50μmol/L人参皂苷Rg1组巨噬细胞内IL-6 mRNA和NEAT1表达下调,培养上清中IL-6分泌减少,细胞存活率上升(均P<0.05),5μmol/L人参皂苷Rg1组则无此现象。结论人参皂苷Rg1可抑制LPS诱导的巨噬细胞炎症反应,其机制可能与下调IL-6 mRNA、NEAT1表达,抑制IL-6蛋白分泌有关。
Objective To test the effect of ginsenoside Rg1 on the expression of inflammatory factor IL-6 and pro-inflammatory factor lncRNA NEAT1 in RAW264.7 macrophages induced by LPS.Methods RAW264.7 macrophages were divided into control group,LPS group,and LPS combined with ginsenoside Rg1 group at different concentrations(5,50μmol/L).Cells and culture supernatant were collected after 6h,respectively.Cell survival rate was detected by MTT,intracellular IL-6 mRNA and NEAT1 expression was detected by RT-qPCR,and cytokine IL-6 expression in supernatant was detected by ELISA.Results Compared with the control group,the expression of IL-6 mRNA and NEAT1 in macrophages was up-regulated by LPS,the IL-6 protein secretion in the culture supernatant increased,and the cell survival rate decreased(all P<0.05).The expression of IL-6 mRNA and NEAT1 in macrophages was down-regulated in the 50μmol/L ginsenoside Rg1 group,the IL-6 secretion in the culture supernatant was decreased,and the cell survival rate was increased(all P<0.05),but no such phenomenon was found in the 5μmol/L ginsenoside Rg1 group.Conclusion Ginsenoside Rg1 can inhibit LPS-induced inflammatory response of macrophages,and its mechanism may be related to the down-regulation of IL-6 mRNA and NEAT1 expression,and the inhibition of IL-6 protein secretion.
