摘要
为进一步了解新城疫病毒(NDV)的遗传进化规律,从疑似NDV感染的鸽群中通过鸡胚接种分离出1株新城疫强毒株,将其命名为Pigeon/China/SHJD13/2017,经噬斑纯化后测定其鸡胚平均死亡时间(MDT)为57.2 h,1日龄雏鸡脑内致病指数(ICPI)为2,毒力指标属于强毒NDV。参考GenBank公布的NDV全基因组序列,共设计了11对引物,对其进行全基因组测序,序列测定结果显示:分离株属于基因Ⅶ型NDV,其基因组全长15192 bp,登录号为MK937786,F基因共包含553个aa,裂解位点的氨基酸残基为112 R-R-Q-K-R-F 117,符合NDV强毒株的分子特征。通过序列同源性比对分析,该分离株与鸡源强毒株Chicken/China/SDWF07/2011亲缘关系较近,表明该鸽源分离毒株可能来自于感染的鸡群。
A strain of Newcastle disease virus(NDV)was isolated from pigeon flocks suspected to be infected with NDV and named as Pigeon/China/SHJD13/2017.After 3 rounds of plaque purification,we detected the pathogenicity assessment indices.The mean time of embryo death(MDT)was 57.2 h,and the intracerebral pathogenicity index(ICPI)was 2,indicating that the isolate was velogenic.With reference to the NDV genome sequences already published in GenBank,11 pairs of primers were designed for full genome amplification.Sequence analysis showed that the genome size of the isolate was 15192 bp(GenBank accession No.MK937786)and phylogenetic analysis indicated that the isolate belonged to genotypeⅦ.The F gene coded 553 amio acids and the cleavage site was 112 R-R-Q-K-R-F 117,which possesses the virulent NDV molecular characteristics.Sequence homology analysis showed that the isolate was closely related to Chicken/China/SDWF07/2011,indicating that the isolate from pigeon might originate from infected chickens.
