摘要
目的评估不同浓度的菟丝子乙醇提取物对宫颈癌Hela细胞增殖和凋亡的影响。方法将培养好的宫颈癌Hela细胞分为5组,对照组仅给予1%DMSO,其余4组为实验组,分别加入3.125、6.25、12.5、25 mg/ml菟丝子乙醇提取物。分别在培养12、24、48 h时检测HeLa细胞增殖情况,培养48 h后分析各组的细胞周期以及细胞迁移和侵袭能力的变化;检测Caspase-3和Bcl-2蛋白的表达。结果与对照组相比,仅12.5、25 mg/ml组G2/M期的细胞数比例显著增加,G0/G1期细胞比例下降(均P<0.05)。培养24、48 h时25 mg/ml组对Hela细胞的增殖抑制率均显著高于其余各组(均P<0.05),12.5 mg/ml组在24、48 h的增殖抑制率均显著高于12 h的增殖抑制率(均P<0.05),25 mg/ml组的增殖抑制率随着时间增加显著增加(均P<0.05)。迁移及侵袭实验中12.5、25 mg/ml组的穿膜细胞数较对照组均明显减少,Hela细胞中Caspase-3表达较对照组显著增加,Bcl-2蛋白表达较对照组显著减少(均P<0.05)。结论高浓度的菟丝子乙醇提取物可以通过阻滞细胞周期于G2/M期,抑制Hela细胞的侵袭和迁移,通过上调Caspase-3和下调Bcl-2促进Hela细胞凋亡,提示高浓度的菟丝子乙醇提取物具有抗宫颈癌的作用。
Objective To evaluate the effects of different concentrations of ethanol extract of cuscutae chinensis on the proliferation,apoptosis and migration of cervical cancer Hela cells.Methods Cervical cancer Hela cells were divided into 5 groups.The control group was given 1%DMSO,and the other 4 groups were treated with semen cuscutae ethanol extracts at different levels(3.125,6.25,12.5,25 mg/ml).The proliferation of Hela cells was detected at 12,24,48 h of culture.The cell cycle,cell migration and invasion ability was analyzed after culture for 48 h,and the expression of Caspase-3 and Bcl-2 protein was also detected.Results Compared with the control group,the proportion of cells in the G2/M phase was significantly increased in 12.5 and 25 mg/ml treatment groups,while the proportion of cells at G0/G1 phase was relatively decreased(all P<0.05).The proliferation inhibition rate of Hela cells in 25 mg/ml treatment group was significantly higher than that in the other groups at 24 and 48 h culture(all P<0.05).The inhibition rate of 12.5 mg/ml group at 24 and 48 h culture was significantly higher than that at 12 h(all P<0.05).The proliferation inhibition rate of 25 mg/ml group increased significantly with the increase of time(all P<0.05).The number of transmembrane cells in 12.5 and 25 mg/ml treatment groups was significantly lower than that in the control group.The expression of Caspase-3 in Hela cells was significantly higher than that in the control group,and the expression of Bcl-2 protein was significantly lower than that in the control group(all P<0.05).Conclusion High concentration of semen cuscutae ethanol extracts can inhibit the invasion and migration of Hela cells by blocking the cell cycle at G2/M stage,and promote the apoptosis of Hela cells by up-regulating caspase-3 and down-regulating Bcl-2,suggesting that high concentration of semen cuscutae extracts may have an anti-cervical cancer effect.
