摘要
目的:建立多西他赛他莫昔芬复方脂质体的包封率测定方法。方法:复方脂质体的制备采用薄膜分散法,用葡聚糖凝胶柱层析法分离脂质体和游离药物,采用高效液相色谱法测定脂质体中多西他赛和他莫昔芬包封率。结果:在选择的色谱条件下,多西他赛和他莫昔芬专属性良好,多西他赛在浓度0.4~10μg/mL,枸橼酸他莫昔芬在浓度1~15μg/mL内线性关系良好,重现性日间日内精密度均符合药典要求。流出曲线实验表明,能有效分离游离药物和脂质体,测定的复方脂质体平均包封率多西他赛(84.42±0.85)%,他莫昔芬(94.78±0.53)%。结论:该测定方法可用于复方脂质体包封率的测定。
Objective:To establish a method to determine the entrapment efficiency of co-delivery of docetaxel(DTX)and tamoxifen(TMX)with Liposome.Method:Co-delivery of DTX and TMX with Liposome are prepared by film dispersion method.Entrapment efficiency is determined after the separation of the unentrapped drug by glucan gel column chromatography.The concentrations of DTX and TMX is determined by HPLC.Result:Under the selected chromatographic conditions,DTX and TMX had good specificities.The concentrations of DTX and TMX had good linear correlations in the ranges of 0.4-10μg/mL and 1-15μg/mL,respectively.The elution profile shows that drug loaded liposomes has a good separation with free drug.The average entrapment efficiency of DTX and TMX in liposomes is(84.42±0.85)%and(94.78±0.53)%.Conclusion:This simultaneous determination by gel filtration combined with HPLC method is dependable is accurate,credible and it can be used to determine the entrapment efficiency of Docetaxel and Tamoxifen in liposomes.
