摘要
目的:探讨MAPK通路蛋白磷酸化酪氨酸磷酸化蛋白激酶(p-p38)、酪氨酸磷酸化蛋白激酶(p38)、磷酸化细胞外信号调节性激酶(p-ERK)、细胞外信号调节性激酶(ERK)在肾透明细胞癌中的表达及意义。方法:收集石河子大学医学院第一附属医院2002-2017年肾透明细胞癌患者90例及正常肾组织患者80例,制备组织芯片2张,免疫组织化学检测p-p38、p38、p-ERK和ERK的表达,结合患者临床病理特征进行分析。结果:(1)p-p38蛋白在肾透明细胞癌(81.7%,58/71)中较正常肾组织(54.5%,36/66)高表达,差异具有统计学意义(P<0.05);(2)p38蛋白在正常肾组织(91.2%,62/68)中较肾透明细胞癌(59.7%,43/72)高表达,差异具有统计学意义(P<0.05);(3)p-ERK蛋白在肾透明细胞癌(60.3%,44/73)中较正常肾组织(31.9%,22/69)高表达,差异具有统计学意义(P<0.05);(4)ERK蛋白在正常肾组织(81.2%,56/69)中表达高于肾透明细胞癌(72.6%,53/73),但无统计学差异(P>0.05)。结论:MAPK信号通路可能参与了肾透明细胞癌的发生发展。
Objective:To investigate the MAPK signaling pathway related protein p-p38(Phosphorylated tyrosine phosphorylated protein kinase,p-p38),p38(Tyrosine phosphorylated protein kinase,p38),p-ERK(Phosphorylated extracellular signaling regulatory kinases,p-ERK)and ERK(Extracellular signaling regulatory kinases,ERK)in differential expression and significance in renal clear cell carcinoma.Methods:90 patients with renal clear cell carcinoma and 80 patients with normal renal tissue were collected from the First Affiliated Hospital of Shihezi University School of Medicine from 2002 to 2017.Two tissue microarrays were prepared.Immunohistochemistry was used to detect p-p38,p38,p-ERK and ERK.The expressions of four biomarkers were combined with the clinical and pathological features of the patients.Results:(1)The expression of p-p38 in renal clear cell carcinoma(81.7%,58/71)was significantly higher than normal renal tissue(54.5%,36/66),P<0.05.(2)The expression of p38 in normal renal tissue(91.2%,62/68)was significantly higher than renal clear cell carcinoma(59.7%,43/72),P<0.05.(3)The expression of p-ERK in renal clear cell carcinoma(60.3%,44/73)was significantly higher than normal renal tissue(31.9%,22/69),P<0.05.(4)The expression of ERK in normal renal tissue(81.2%,56/69)was higher than renal clear cell carcinoma(72.6%,53/73),but P>0.05.Conclusion:The MAPK signaling pathway may be involved in the development of renal clear cell carcinoma.
