摘要
目的观察补肾生髓强骨方对酒精性股骨头坏死模型大鼠炎症因子、氧化应激及toll样受体4(TLR4)、核因子-κB(NF-κB)和血管内皮生长因子(VEGF)蛋白表达的影响。方法将30只烈性白酒诱导的酒精性股骨头坏死模型大鼠随机分为模型组、干预组和阳性组,另选择10只Wistar大鼠作为对照组,模型组和对照组给予生理盐水灌胃,干预组给予补肾生髓强骨汤灌胃,阳性组给予美多芭灌胃,剂量均为10 mL/kg,连续给药4周。HE染色比较各组大鼠的骨组织的病理学改变,比较炎症因子、氧化应激指标,骨组织中TLR4、NF-κB和VEGF基因和蛋白表达。结果模型组股骨头组织结构紊乱,软骨脱落,骨细胞陷窝,脂肪细胞体积增大;干预组和阳性组股骨头组织结构改善,软骨少量脱落,骨细胞陷窝减轻,脂肪细胞体积增大不明显。干预组和阳性组血清TNF-α、IL-6和CRP水平显著降低,且干预组低于阳性组(P<0.05)。干预组和阳性组脂质过氧化物(LPO)、丙二醛(MDA)水平降低,超氧化物歧化酶(SOD)和谷胱甘肽过氧化酶(GSH-PX)水平升高,且干预组优于模型组(P<0.05)。干预组和阳性组TLR4、NF-κB基因和蛋白表达水平降低,VEGF基因和蛋白表达水平升高,且干预组优于阳性组(P<0.05)。结论补肾生髓强骨方对酒精性股骨头坏死模型大鼠具有较好的治疗作用,可抑制炎症因子,改善氧化应激,其机制可能与抑制TLR4、NF-κB和VEGF蛋白表达有关。
Objective:To investigate the effects of Bushen Shengsui Qianggu Decoction on inflammatory factors,oxidative stress,and protein expression of toll-like receptor 4(TLR4),nuclear factor κB(NF-κB) and vascular endothelial growth factor(VEGF) in alcoholic femoral head necrosis model rats.Methods:A total of 30 alcohol-induced femoral head necrosis model rats induced by strong liquor were randomly divided into the model group,intervention group and positive group.Another 10 Wistar rats were selected as the control group.The model group and the control group were given saline gavage while the intervention group was given Bushen Shengsui Qianggu Decoction by gavage,and the positive group was given Metopa gavage at the dose of 10 mL/kg for 4 weeks.HE staining was applied to compare the pathological changes of the bone tissue of each group.The inflammatory factors,oxidative stress indicators,TLR4,NF-κB and VEGF gene and protein expression were also compared.Results:The tissue structure of the femoral head in the model group was disordered,cartilage fell off,had bone cell lacuna,and increased fat cell volume;the femoral head tissue structure in the intervention group and positive group improved,with a small amount of cartilage falling off,decreased bone cell lacuna,and fat cell volume not obviously increased.The serum TNF-α,IL-6 and CRP levels of the intervention group and the positive group were significantly reduced,and the intervention group were lower than those of the positive group(P <0.05).The LPO and MDA levels of the intervention group and the positive group decreased,and the SOD and GSH-PX levels increased,and the intervention group were better than those of the model group(P <0.05).The expression level of TLR4,NF-κB gene and protein in the intervention group and the positive group decreased,and the expression of VEGF gene and protein increased,and the levels in the intervention group were better than those of the positive group(P <0.05).Conclusion:Bushen Shengsui Qianggu Decoction has good therapeutic effect on alcohol-induced femoral head necrosis in rats.It could reduce inflammatory factors and improve oxidative stress.The mechanism may be related to the inhibition of TLR4,NF-κB and VEGF protein expression.
