丙泊酚对创伤性脑损伤大鼠神经细胞自噬的影响 被引量:1

Effects of propofol on autophagy of neurons in rats with traumatic brain injury

摘要 目的探讨丙泊酚对创伤性脑损伤(TBI)大鼠神经细胞自噬的影响。方法选取雄性健康SPF级SD大鼠72只,依据随机数字表法分为3组:假手术组、TBI模型对照组、TBI+丙泊酚组,每组各24只,再分为3个亚组,每亚组8只。采用自由落体砝码撞击SD大鼠,建立TBI模型,并观察脑组织形态学,检测脑组织细胞凋亡并计算皮质神经元细胞凋亡率,检测自噬相关蛋白(Beclin-1蛋白、P62蛋白及微管相关蛋白1轻链3-Ⅱ/Ⅰ(LC3-II/Ⅰ)蛋白比值)水平。结果TBI模型对照组可见其损伤脑组织区结构混乱无序、神经胶质疤痕明显,疤痕组织显著萎缩且可见软化灶;TBI+丙泊酚组损伤脑组织区有神经胶质细胞填充,其间疤痕组织范围及软化灶区域面积与TBI模型对照组对比均较小。假手术组、TBI模型对照组、TBI+丙泊酚组皮质神经元细胞凋亡率分别为(9.78±0.69)%、(67.39±2.57)%、(30.73±2.06)%,假手术组显著低于TBI模型对照组及TBI+丙泊酚组,TBI+丙泊酚组显著低于TBI模型对照组(P<0.05)。TBI+丙泊酚组Beclin-1蛋白及LC3-Ⅱ/Ⅰ蛋白比值均显著低于TBI模型对照组,P62蛋白表达高于TBI模型对照组(P<0.05)。结论丙泊酚对TBI大鼠神经细胞具有一定程度的保护作用,其机制包括对受损神经细胞凋亡及自噬的抑制。 Objective To investigate the effect of propofol on autophagy of nerve cells in rats with traumatic brain injury(TBI).Methods 72 male healthy SD rats were divided into three groups according to the randomly number table method:sham operation group,TBI model control group and TBI-propofol group(n=24),and then each group were divided into three subgroups group,8 in each subgroup.The TBI model was established by impacting SD rats with free falling weight,the morphology of brain tissue were observed,the brain tissue cell apoptosis were detected and the apoptosis rate of cortical neurons were calculate,the autophagy-related proteins(Beclin-1 protein,P62 protein)and microtubule-associated protein 1 light chain 3-Ⅱ/Ⅰ(LC3-Ⅱ/Ⅰ)protein ratio levels were detected.Results In the TBI model control group,the structure of the injured brain tissue was disordered,the glial scar was obvious,the scar tissue was significantly atrophy and softened;Compared with the TBI model control group,the tissue range and softening area were smaller in the injured brain tissue in the TBI-propofol group which was filled with glial cells and there were scars in the middle.The apoptosis rates of cortical neurons in the sham operation group,TBI model control group,and TBI-propofol group were respectively(9.78±0.69)%,(67.39±2.57)%and(30.73±2.06)%.The sham operation group was significantly lower than TBI model control group and TBI-propofol group,TBI-propofol group was significantly lower than TBI model control group(P<0.05).The ratio of Beclin-1 protein and LC3-Ⅱ/Ⅰ protein in the TBI-propofol group was significantly lower than the TBI model control group,and the expression of P62 protein was higher than the TBI model control group(P<0.05).Conclusion Propofol has a certain degree of protective effect on nerve cells of TBI rats.The mechanism includes inhibition of apoptosis and autophagy of damaged nerve cells.
