摘要
目的探讨温针灸及骨髓间充质干细胞(BMSCs)移植对膝骨性关节炎(KOA)模型兔功能的影响。方法将50只新西兰兔随机分为空白组(CT组)、模型组(Model组)、温针灸组(WNM组)、骨髓间充质干细胞组(BMSCs组)、温针灸联合骨髓间充质干细胞组(WNM+BMSCs组)。采用右后肢伸直位固定法制备兔KOA中期软骨退变模型,依据Mankin法予以评估软骨组织损伤程度;用ELISA法检测MMP-1、MMP-13和IL-10的含量表达;采用Western blot检测MMP-1、MMP-13蛋白表达。结果Model组Mankin评分高于CT组(P<0.05);治疗后WNM组、BMSCs组、WNM+BMSCs组Mankin评分均比MX组低(P<0.05),但比CT组升高(P<0.05)。WNM+BMSCs组Mankin评分低于WNM组与BMSCs组,差异有统计学意义(P<0.05);经温针灸与BMSCs移植治疗后,兔KOA模型血清中MMP-1和MMP-13均下降,IL-10水平升高,且WNM+BMSCs组治疗效果优于BMSCs组及WNM组(P<0.05);WNM+BMSCs组MMP-1、MMP-13蛋白水平显著降低。结论温针灸与BMSCs移植可以降低MMP-1和MMP-13水平,升高IL-10水平的表达。
Objective To investigate the effects of warming needle moxibustion combined with bone marrow mesenchymal stem cells(BMSCs)transplantation on knee osteoarthritis(KOA)model in rabbits.Methods Fifty New Zealand rabbits were randomly divided into blank group(CT),model group(Model),Warming Needle Moxibustion group(WNM),bone marrow mesenchymal stem cell group(BMSCs group),Warming Needle Moxibustion combined with bone marrow mesenchymal stem cell group(WNM+BMSCs).The middle stage cartilage degeneration model of KOA was established by right hind limb extension fixation method,and the degree of the injured cartilage was evaluated according to the Mankin method.The expression of MMP-1,MMP-13 and IL-10 were measured by ELISA.The protein expression of MMP-1,MMP-13 were detected by Western blotting.Results The Mankin score of the model group was higher than that of the CT group(P<0.05).After treatment,the Mankin scores of the WNM group,BMSCs group,and WNM+BMSCs group were lower than those of the MX group(P<0.05),but higher than those of the CT group(P<0.05).The Mankin score of the WNM+BMSCs group was lower than that of the WNM and BMSCs groups(P<0.05),which was statistically significant.After Warming Needle Moxibustion and BMSCs transplantation,MMP-1 and MMP-13 in the serum of rabbit KOA models decreased,and the level of IL-10 level increased,and the therapeutic effect of WNM+BMSCs group was better than that of BMSCs group and WNM group(P<0.05).The protein levels of MMP-1 and MMP-13 in the WNM-BMSCs group were significantly reduced.Conclusion Warming Needle Moxibustion and BMSCs transplantation can reduce the levels of MMP-1 and MMP-13 and increase the expression of IL-10.
