摘要
为了研究枸杞多糖及其羧甲基化衍生物的结构和抗氧化性能,使用微波辅助酶解法提取枸杞多糖并进行羧甲基化修饰。采用高效液相色谱仪(HPLC)、傅里叶变换红外光谱仪(FTIR)和尺寸排阻色谱-多角度激光光散射-示差折光联用仪(SEC-MALLS-RI),对枸杞多糖进行了结构表征,对枸杞多糖及其羧甲基化衍生物的抗氧化性能进行了测定。研究结果表明:枸杞多糖的提取率为4.32%(质量分数),羧甲基化枸杞多糖取代度为0.43。枸杞多糖主要由甘露糖(40.05%,摩尔分数,下同)、葡萄糖醛酸(18.98%)、阿拉伯糖(18.34%)、鼠李糖(15.73%)和葡萄糖(6.90%)组成。枸杞多糖的数均相对分子质量为3.561×10^4,质均相对分子质量为5.059×10^4,多分散系数为1.421,均方根旋转半径为73.0 nm。在相同质量浓度下,羧甲基化枸杞多糖的抗氧化性能高于枸杞多糖。
In order to study the structure and antioxidant activities of Lycium barbarum(L.barbarum)polysaccharide and its carboxymethylation derivative,the L.barbarum polysaccharide was extracted by microwave-assisted enzymatic hydrolysis method and was modified by carboxymethylation.The structure of L.barbarum polysaccharide was characterized by high performance liquid chromatograph(HPLC),Fourier transform infrared spectrometer(FT-IR)and size exclusion chromatography-multi-angle laser light scattering-refractive index(SEC-MALLS-RI).The antioxidant activities of L.barbarum polysaccharide and its carboxymethylation derivative were determined.The results show that the extraction rate of L.barbarum polysaccharide is 4.32%(mass fraction),and the substitution degree of carboxymethylated L.barbarum polysaccharide is 0.43.The L.barbarum polysaccharide is mainly composed of mannose(40.05%,mole fraction,the same below),glucuronic acid(18.98%),arabinose(18.34%),rhamnose(15.73%)and glucose(6.90%).The number-average relative molecular weight of L.barbarum polysaccharide is 3.561×10^4,the mass-average relative molecular weight is 5.059×10^4,the polydispersity coefficient is 1.421,and the root mean square rotation radius is 73.0 nm.At the same mass concentration,the antioxidant activities of carboxymethylated L.barbarum polysaccharide are higher than that of L.barbarum polysaccharide.
