流动注射-光度法自动测定多种植物样品中过氧化物酶活性 被引量:2

Automatic determination of peroxidase activity in various plants by flow injection-spectrophotometry

摘要 基于愈创木酚(GA)/过氧化物酶(POD)/过氧化氢(H2O2)反应体系,建立一种新的流动注射分析流路,测定多种植物样品中过氧化物酶活性。结果表明,系统参数优化如下:系统流速1.91 mL/min,样品进样体积40μL,试剂注入体积150μL,反应盘管长度150 cm,磷酸盐缓冲溶液浓度0.05 mol/L,pH 6.0;HCl清洗液进样体积150μL,浓度0.24 mol/L;底物为4.5 mmol/L GA和1.5 mmol/L H2O2混合溶液;反应产物的表观摩尔吸光系数测定结果为0.0064 L/(μmol·cm)。本方法测定范围在153~2752 U/L之间,检出限为25.7 U/L,重现性较好(RSD≤3.38%,n=11),分析速度为90样/h。对五种植物样品中的过氧化物酶活性测定并进行加标回收实验,回收率在95%~105%之间。 Based on guaiacol(GA)/peroxidase(POD)/hydrogen peroxide(H2O2) reaction system,a new FIA analytical flow path for determination of POD activity in various plant samples was established and the parameters of the system were optimized.The results showed that the optimized parameters were as follows:the flow rate of the system was 1.91 mL/min,the sample injection volume was 40 μL,the reagent injection volume was 150 μL,the length of the reaction coil was 150 cm,the concentration of phosphate buffer solution was 0.05 mol/L,pH 6.0;HCl cleaning agent injection volume was 150 μL,the concentration was 0.24 mol/L;the reaction reagent was the mixture of 4.5 mmol/L GA and 1.5 mmol/L H2O2;the apparent molar absorption coefficient of the product was 0.006 4 L/(μmol·cm).The reproducibility of this method was superior(RSD≤3.38%,n=11),the determination range was between 153~2 752 U/L with detection limit of 25.7 U/L,the analysis speed was 90 samples/h.The POD in five kinds of plant samples was determined and the recovery rate was between 95%~105%.
