摘要
目的:观察针刺对帕金森抑郁(Parkinson′s disease with depression,PDD)模型的作用及机制。方法:SD大鼠随机分为正常组、假手术组、模型组、对照[氟西汀(6 mg·kg^-1)+多巴丝肼片(6 mg·kg^-1)]组、针刺组,每组15只。采用单侧纹状体双靶点注射法和慢性不可预见性温和性应激法制备PDD模型。针刺组针刺"百会透太阳""中脘",每日治疗20 min,其他组给予相应的药物灌胃。各组大鼠在28 d后测体质量变化率,蔗糖溶液偏好和旷场实验;酶联免疫(ELISA)法检测大鼠血清脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)的水平;HE染色观察脑病理变化;免疫组化法检测大鼠脑组织BDNF表达。结果:与正常组比较,模型组大鼠出现明显的帕金森伴抑郁样行为,脑组织BDNF的表达及血清BDNF水平明显降低(P<0.05);与模型组相比,针刺组和对照组的帕金森伴抑郁样行为有明显改善,同时脑组织BDNF的表达及血清BDNF水平明显升高(P<0.05),针刺组和对照组组间比较无统计学差异(P>0.05)。结论:针刺对PDD模型大鼠的帕金森伴抑郁样行为有明显的改善作用,并能提高其脑组织及血清BDNF的表达水平,从而发挥治疗作用。
Objective:To observe the effect and mechanism of acupuncture on model rates with Parkinson′s disease with depression(PDD).Methods:SD rats were randomly divided into normal group,sham operation group,model group,control group[fluoxetine(6 mg·kg^-1)+dopaserazine tablets(6 mg·kg^-1)],acupuncture group,15 per group.The PDD model was prepared by two-spot unilateral striatal injection and chronic unpredictable mild stress(CUMS)method.In the acupuncture group,"Baihui tou Taiyang"and"Zhongwan"acupoints were treated with needling for 20 minutes a day.The other groups were given corresponding drugs by gavage.After 28 days,rats in each group were tested for body weight change rate,sucrose consumption preference,and open-field test(OFT);Enzyme-linked immunoassay(ELISA)method was used to detect the level of serum brain-derived neurotrophic factor(BDNF)in SD rats.Hematoxylin-Eosin Staining was used to observe the brain pathological changes and the immunohistochemical method was used to detect the secretion level of BDNF in rat brain tissue.Results:Compared with the normal group,the rats in the model group showed obvious PPD-like behavior,and the secretion of BDNF in brain tissue and the level of serum BDNF were significantly reduced(P<0.05);Compared with the model group,however,the acupuncture group and the control group ameliorated notably in PDD behavior.At the same time,the secretion of BDNF in brain tissue and the level of serum BDNF increased significantly(P<0.05).And there was no statistical difference(P>0.05)between the acupuncture group and the control group.Conclusion:Acupuncture therapy can exert a therapeutic effect by significantly improve Parkinson′s depression-like behavior in PDD model rats and increase the secretion level of BDNF in brain tissue and serum.
