摘要
目的探讨异位病毒整合位点-1(EVI-1)在胃癌中的表达及临床意义。方法采用免疫组织化学法检测胃癌组织和癌旁组织中EVI-1蛋白的表达情况;采用蛋白质印迹法(Western blot)和实时荧光定量逆转录聚合酶链反应,分别检测人胃癌细胞株AGS、NCI-N87、MGC-803、HGC27和永生化正常胃黏膜细胞株GES-1中EVI-1的3个剪切变异体蛋白(MDS1/EVI-1、EVI-1、EVI-1△324)和EVI-1 mRNA(除MGC-803)的表达水平。结果胃癌组织中EVI-1蛋白的阳性表达率为76.7%(46/60),明显高于癌旁正常胃组织的41.7%(25/60),差异有统计学意义(P﹤0.01)。胃癌细胞中MDS1/EVI-1和EVI-1蛋白相对表达量均高于正常胃黏膜上皮细胞,差异均有统计学意义(P﹤0.05);MGC-803、AGS胃癌细胞中EVI-1△324蛋白相对表达量也高于正常胃黏膜上皮细胞,但差异无统计学意义(P﹥0.05)。HGC27、NCI-N87、AGS细胞株中EVI-1 mRNA相对表达量均高于GES-1细胞株,差异均有统计学意义(P﹤0.05)。结论胃癌中EVI-1蛋白(除EVI-1△324亚型)和EVI-1 mRNA的表达均较癌旁正常胃黏膜组织高。EVI-1基因可能是潜在的胃癌原癌基因,尚有待进一步深入研究。
Objective To investigate the expression of ecotropic viral integration site-1(EVI-1)in gastric cancer and its clinical significance.Method The expression of EVI-1 protein in gastric cancer tissues and its adjacent normal tissues were detected by immunohistochemistry(IHC).The expression of EVI-1 splice variant proteins(MDS1/EVI-1、EVI-1 and EVI-1△324)and EVI-1 mRNA in both four gastric cancer cell lines(AGS,NCI-N87,MGC-803,HGC27)and normal gastric mucosal immortalized cells(GES-1)were detected by Western blot and real-time quantitative reverse transcription-polymerase chain reaction(qRT-PCR),respectively.Result At the histological level,the positive expression rate of EVI-1 in gastric cancer tissues was higher than that in adjacent normal tissues[76.7%(46/60)vs 41.7%(25/60),P<0.01].The relative expression levels of MDS1/EVI-1 and EVI-1 in gastric cancer cells were higher than those in normal gastric epithelial cells(P<0.05).The relative expression of EVI-1△324 protein in MGC-803 and AGS gastric cancer cells were also higher than that in normal gastric epithelial cells,but the difference was not statistically significant(P>0.05).The relative expression levels of EVI-1 mRNA in HGC27,NCI-N87 and AGS cell lines were significantly higher than those in GES-1 cell lines(P<0.05).Conclusion The expression of EVI-1 protein(except for EVI-1△324 subtype)and EVI-1 mRNA in gastric cancer were higher than those in adjacent normal gastric mucosa.EVI-1 may be a potential proto-oncogene which is needed to be further studied.
