摘要
目的观察益肾化浊方及拆方对7月龄SAMP8小鼠海马神经元凋亡的影响。方法将40只7月龄SAMP8小鼠随机分成SAMP8(P8)组、益肾化浊方(YSHZ)组,益肾(YS)组和化浊(HZ)组,每组10只,选择10只抗快速老化小鼠(SAMR1)作为对照(R1)组;益肾化浊方、益肾方与化浊方分别按照6.24、4.68、1.56 g/(kg·d)灌胃干预,R1和P8组予等量蒸馏水灌胃,疗程均为4 w,4 w后停止干预。采用TUNEL染色观察各组小鼠海马神经元凋亡指数变化,采用Western印迹检测凋亡途径中关键蛋白B淋巴细胞瘤相关X蛋白(Bax)/B淋巴细胞瘤(Bcl)2、半胱氨酸天冬氨酸特异性蛋白酶(Caspase)9、Caspase10、裂解(Cleaved)Caspase3表达情况。结果海马CA1区神经元凋亡指数:与R1组比较,P8组凋亡指数明显增高(P<0.05)。与P8组比较,YSHZ组凋亡指数明显降低(P<0.05)。Western印迹检测结果:与R1组相比,P8组小鼠海马促凋亡蛋白Bax/Bcl2比值、Caspase9、Caspase10和Cleaved Caspase3表达明显升高(P<0.05)。与P8组相比,YSHZ组Bax/Bcl2比值、Caspase10和Cleaved Caspase3表达明显降低(P<0.05);YS组Caspase10表达明显降低(P<0.05)。结论益肾化浊方能明显减轻SAMP8小鼠海马神经元凋亡,该作用可能与调节Bcl-2家族蛋白表达,减少Caspase家族蛋白的活化有关;拆方中益肾方中药具有抗凋亡的作用趋势。
Objective To investigate the effects of YishenHuazhuo prescription and its disassembled prescription on neuron apoptosis in hippocampus of SAMP 8 mice.Methods Forty 7-month-old SAMP8 mice were randomly divided into model(P8)group,YishenHuazhuo(YSHZ)group,Yishen(YS)group,and Huazhuo(HZ)group,10 mice in each group.Ten 7-month-old SAMR1 mice were used as control(R1)group.The YSHZ,YS and HZ mice were treated with YishenHuazhuo prescription,Yishen prescription and Huazhuo prescription by intragastric administration.The corresponding dosages were 6.24,4.68 and 1.56 g/(kg·d),respectively.R1 and P8 groups were given equal distilled water for 4 weeks.After 4 weeks,the intervention was stopped.TUNEL staining and Western blot were used to detect the hippocampal neurons apoptosis and expression of related proteins(Bax/Bcl-2,Caspase9,Caspase10,Cleaved Caspase3)in apoptotic pathways in each group.Results Apoptosis index of hippocampal CA1 neurons:compared with R1 group,the apoptosis index of P8 group was significantly higher(P<0.05);compared with P8 group,the apoptosis index of YSHZ group was significantly lower(P<0.05).Western blot results:compared with R1 group,the Bax/Bcl-2 ratio,Caspase9,Caspase10,and Cleaved Caspase3 expression in the hippocampus of the P8 group were significantly higher(P<0.05);compared with P8 group,Bax/Bcl-2 ratio,Caspase10 and Cleaved Caspase3 expression in YSHZ group were significantly lower(P<0.05);Caspase10 expression significantly decreased in YS group(P<0.05).Conclusions The mechanism of YishenHuazhuo prescription reducing the apoptosis of neurons might be related to regulating of Bcl-2 proteins family,and reducing the activation Caspase proteins family.Chinese herbs in Yishen prescription might have the anti-apoptosis function.
