摘要
目的观察胰岛素样生长因子1受体长链非编码RNA(IRAIN)在食管鳞状细胞癌(ESCC)组织中的表达变化,并探讨其启动子甲基化状态与患者临床病理参数的关系。方法行手术治疗的ESCC患者63例,术中取肿瘤组织和癌旁正常组织,分别采用QPCR法和MSP法检测组织中IRAIN和启动子甲基化状态,并分析IRAIN启动子甲基化状态与IRAIN相对表达量和患者临床病理参数的关系。结果ESCC组织中IRAIN相对表达量低于癌旁正常组织(0.816±0.286比1.000±0.185,P<0.05)。IRAIN启动子甲基化的ESCC组织中IRAIN相对表达量、IRAIN高表达率均低于非甲基化的ESCC组织(P均<0.05)。ESCC组织中IRAIN启动子甲基化状态与患者肿瘤最大直径、TNM分期、淋巴结转移有关(P均<0.05)。结论ESCC组织中IRAIN表达降低、启动子甲基化率升高,IRAIN表达降低可能与其启动子甲基化状态有关;IRAIN启动子甲基化率与肿瘤转移有关。
Objective To observe the expression of insulin-like growth factor 1 receptor antisense imprinted long non-coding RNA(IRAIN)in the esophageal squamous cell carcinoma(ESCC)tissues and to explore the relationship be⁃tween the promoter methylation status of IRAIN and the clinicopathological features.Methods In 63 ESCC patients who underwent surgical resection,tumor tissues and adjacent normal tissues were taken during the operation,and the expres⁃sion and promoter methylation status of IRAIN were detected by QPCR and MSP methods,respectively.The relationships between the promoter methylation status of IRAIN and the relative expression of IRAIN and the clinicopathological charac⁃teristics of patients were analyzed.Results The relative expression of IRAIN in the ESCC tissues was lower than that in the adjacent normal tissues[68.25%(43/63)vs.30.16%(19/63),P<0.05].The relative expression of IRAIN and the high expression rate of IRAIN in ESCC tissues with methylated IRAIN gene promoter were lower than those in non-methyl⁃ated ESCC tissues(all P<0.05).The promoter methylation status of IRAIN gene in the ESCC tissues was related to the maximal tumor diameter,TNM stage,and lymph node metastasis(all P<0.05).Conclusions In ESCC tissues,the ex⁃pression of IRAIN decreases,while the promoter methylation rate of IRAIN gene increases.The down-regulation of IRAIN may be related to the promoter methylation status of IRAIN gene,and the promoter methylation rate of IRAIN gene may be related to the tumor metastasis.
