摘要
目的检测长链非编码RNA肿瘤易感候选者9(long non-coding RNA cancer susceptibility candidate 9,lncRNA CASC9)、微小RNA-193a-5p(microRNA-193a-5p,miR-193a-5p)在子宫内膜癌组织中的表达水平,并探讨在子宫内膜癌组织中两者表达的意义及其相关性。方法starBase v2.0在线软件预测lncRNA CASC9与miR-193a-5p的结合位点;收集我院2017年6月至2019年6月期间诊治的子宫内膜癌组织80例和正常子宫内膜组织40例,采用荧光定量PCR(quantitative real-time PCR,qRT-PCR)检测lncRNA CASC9及miR-193a-5p在子宫内膜癌和正常子宫内膜组织中的表达水平;Spearman相关性分析子宫内膜癌组织中lncRNA CASC9与miR-193a-5p表达的相关性;t检验分析lncRNA CASC9及miR-193a-5p的表达水平与子宫内膜癌患者临床病理参数的关系;Kaplan-Meier生存曲线分析lncRNA CASC9及miR-193a-5p的表达水平对子宫内膜癌患者预后的影响;Cox单因素和多因素回归分析影响子宫内膜癌患者独立预后危险因素。结果lncRNA CASC9与miR-193a-5p存在结合位点。与正常子宫内膜组织相比,子宫内膜癌组织中lncRNA CASC9的表达增高(P<0.05),miR-193a-5p的表达降低(P<0.05)。子宫内膜癌组织中lncRNA CASC9与miR-193a-5p的表达呈负相关(P<0.05)。lncRNA CASC9的表达与子宫内膜癌患者的淋巴结转移、FIGO分期晚期和肌层浸润相关(P<0.05)。miR-193a-5p的表达与子宫内膜癌患者的淋巴结转移和FIGO分期晚期相关(P<0.05)。lncRNA CASC9高表达的子宫内膜癌患者预后差(P<0.05),miR-193a-5p低表达的子宫内膜癌患者预后差(P<0.05)。lncRNA CASC9高表达、miR-193a-5p低表达、淋巴结转移、肌层浸润和FIGO分期晚期是子宫内膜癌患者不良预后的独立危险因素。结论lncRNA CASC9高表达和miR-193a-5p低表达均与子宫内膜癌患者的不良病理参数和预后相关。lncRNA CASC9、miR-193a-5p可能是子宫内膜癌潜在预后标志物和治疗靶点。
Objective To detect the expressions of long non-coding RNA cancer susceptibility candidate 9(lncRNA CASC9)and microRNA-193a-5p(miR-193a-5p)in endometrial cancer tissues,and to explore the significance and correlation of their expressions in endometrial cancer tissues.Methods StarBase v2.0 online software was used to predict the binding sites of lncRNA CASC9 and miR-193a-5p.We collected 80 cases of endometrial cancer tissues and 40 cases of normal endometrial tissues which were diagnosed and treated in our hospital from June,2017 to June,2019.Quantitative real-time PCR(qRT-PCR)was used to detect the expression levels of lncRNA CASC9 and miR-193a-5p in endometrial cancer and normal endometrial tissues.Spearman correlation was used to analyze the correlation between lncRNA CASC9 and miR-193a-5p expressions in endometrial cancer tissues and t test was used to analyze the relationship between the expression levels of lncRNA CASC9 and miR-193a-5p and clinical pathological parameters of patients with endometrial cancer.Kaplan-Meier survival curve was applied to analyze the effect of lncRNA CASC9 and miR-193a-5p expressions on the prognosis of endometrial cancer patients.Cox univariate and multivariate regression analysis were used to analyze the independent prognostic risk factors affecting the prognosis of endometrial cancer patients.Results lncRNA CASC9 and miR-193a-5p showed binding sites through our results.Compared with normal endometrial tissue,the expression of lncRNA CASC9 in endometrial cancer tissue was increased(P<0.05),while the expression of miR-193a-5p was decreased(P<0.05).The expression of lncRNA CASC9 in endometrial cancer tissue was negatively correlated with the expression of miR-193a-5p(P<0.05).The expression of lncRNA CASC9 was associated with lymph node metastasis,FIGO advanced stage and muscle layer infiltration in patients with endometrial cancer(P<0.05).The expression of miR-193a-5p was related to the lymph node metastasis and FIGO advanced stage in patients with endometrial cancer(P<0.05).The prognosis of patients with a high expression of lncRNA CASC9 was poor(P<0.05),and prognosis of patients with a low expression of miR-193a-5p was poor(P<0.05).A high expression of lncRNA CASC9,low expression of miR-193a-5p,lymph node metastasis,muscle layer infiltration and FIGO advanced stage were all independent risk factors for a poor prognosis in patients with endometrial cancer.Conclusion The high expression of lncRNA CASC9 and low expression of miR-193a-5p are related to the adverse pathological parameters and prognosis of endometrial cancer patients.lncRNA CASC9 and miR-193a-5p could serve as potential prognostic markers and therapeutic targets for endometrial cancer.
