小青龙汤对肺癌大鼠VEGF/VEGFR信号通路的影响研究 被引量:8

摘要 目的基于VEGF/VEGFR信号通路探讨小青龙汤对肺癌大鼠的作用及机制。方法取24只成年雄性SD大鼠,利用尾静脉注射含Walker-256细胞的腹水细胞悬液法建立肺癌模型,造模完成后随机分为模型组、小青龙汤组、贝伐珠单抗组、小青龙汤联合贝伐珠单抗组各6只,另取6只大鼠不做任何处理作为对照组。小青龙汤组给予小青龙汤10 mL/kg灌胃,贝伐珠单抗组给予贝伐珠单抗2 mg/kg腹腔注射,小青龙汤联合贝伐珠单抗组给予小青龙汤10 mL/kg灌胃及贝伐珠单抗2 mg/kg腹腔注射,模型组及对照组灌胃给予等量生理盐水,均1次/d,连续28 d。实时定量PCR法检测各组大鼠肺组织中p16、p21、p53 mRNA表达情况,流式细胞术检测肺组织细胞凋亡率,苏木精-伊红染色法观察肺组织病理学变化,Western blot法检测肺组织中VEGF、VEGFR2表达水平。结果与模型组比较,小青龙汤组、贝伐珠单抗组及小青龙汤联合贝伐珠单抗组大鼠肺组织中p16、p21、p53 mRNA相对表达量和VEGF、VEGFR2表达水平均显著降低(P均<0.05),肺细胞凋亡率均显著升高(P均<0.05),肺组织病理学明显改善,且小青龙汤联合贝伐珠单抗组较其他组变化更明显(P均<0.05)。结论小青龙汤能够促进大鼠肺癌细胞凋亡,改善肺组织病理学变化,其机制可能与调节VEGF/VEGFR信号通路有关。 Objective It is to study the effect and mechanism of Xiaoqinglong Decoction on lung cancer in rats based on the VEGF/VEGFR signal pathway.Methods Twenty-four adult male SD rats were selected and given the ascites cell suspension containing Walker-256 cells by injection into the tail vein to establish lung cancer models.After the modeling was completed,they were randomly divided into model group,Xiaoqinglong Decoction group,bevacizumab group and Xiaoqinglong Decoction plus bevacizumab group(combination group),each group had 6 rats,another 6 rats without any treatment as the control group.Xiaoqinglong Decoction group was given Xiaoqinglong Decoction 10mL/kg by gavage,bevacizumab group was given bevacizumab 2 mg/kg by intraperitoneal injection,the combiantion group was given Xiaoqinglong Decoction 10 mL/kg by gavage and bevacizumab 2 mg/kg by intraperitoneal injection,and the model group and the control group were given the same amount of normal saline by gavage,all the groups were treated once a day for 28 consecutive days.Real-time quantitative PCR method was used to detect the expression of p16,p21,and p53 mRNA in the lung tissue of rats in each group,flow cytometry was used to detect the apoptosis rate of lung tissue,hematoxylin-eosin staining method was used to observe the pathological changes of lung tissue,Western blot method was used to detect the expression levels of VEGF and VEGFR2 in lung tissue.Results Compared with the model group,the relative expression levels of p16,p21,and p53 mRNA and the expression levels of VEGF and VEGFR2 in the lung tissues of rats in the Xiaoqinglong Decoction group,bevacizumab group and the combination group were significant decreased(P<0.05),lung cell apoptosis rate was significantly increased(P<0.05),lung histopathology was significantly improved,and the changes in the combination group were more significantly than those in the other groups(P all<0.05).Conclusion Xiaoqinglong Decoction can promote the apoptosis of lung cancer cells in rats and improve the pathological changes of lung tissue.The mechanism may be related to the regulation of VEGF/VEGFR signaling pathway.
