摘要
目的探讨肝细胞肝癌(以下简称肝癌)组织中lncRNA NCK1-AS1表达及临床意义。方法选取2015年4月至2017年6月该院肝癌确诊患者73例,采集肝癌组织及配对的癌旁组织,采用实时荧光定量聚合酶链式反应(qRT-PCR)检测lncRNA NCK1-AS1表达水平,分析lncRNA NCK1-AS1与miR-361-5p表达的相关性。根据肝癌组织中lncRNA NCK1-AS1表达水平将研究对象分为高表达组(33例)和低表达组(40例),比较2组临床病理参数及预后。结果肝癌组织中lncRNA NCK1-AS1相对表达量高于癌旁组织,差异有统计学意义(P<0.05)。低表达组分化程度、淋巴转移及TNM分期情况与高表达组比较,差异均有统计学意义(P<0.05)。低表达组中位生存期明显高于高表达组,差异有统计学意义(P<0.05)。肝癌组织中miR-361-5p相对表达量低于癌旁组织,差异有统计学意义(P<0.05)。lncRNA NCK1-AS1与miR-361-5p表达呈负相关(r 2=0.0786,P<0.001)。结论肝癌组织中lncRNA NCK1-AS1呈高表达,与患者不良预后相关,其可能通过调控miR-361-5p表达来发挥作用。
Objective To investigate the expression and clinical significance of lncRNA NCK1-AS1 in hepatocellular carcinoma(HCC)tissues.Methods From April 2015 to June 2017,73 patients with HCC in the hospital were selected.HCC tissues and matched adjacent tissues were collected.The expression level of lncRNA NCK1-AS1 was detected by real-time fluorescent quantitative polymerase chain reaction(qRT-PCR),and the correlation between lncRNA NCK1-AS1 and miR-361-5p expression was analyzed.According to the expression level of lncRNA NCK1-AS1 in HCC tissues,the subjects were divided into high expression group(33 cases)and low expression group(40 cases).The clinicopathological parameters and prognosis of the two groups were compared.Results The relative expression of lncRNA NCK1-AS1 in HCC tissues was higher than that in adjacent tissues(P<0.05).The differentiation degree,lymph node metastasis and TNM stage of the low expression group were significantly different from those of the high expression group(P<0.05).The median survival time of low expression group was significantly higher than that of high expression group,and the difference was statistically significant(P<0.05).The relative expression of miR-361-5p in HCC tissues was lower than that in adjacent tissues(P<0.05).lncRNA NCK1-AS1 was negatively correlated with miR-361-5p expression(r 2=0.0786,P<0.001).Conclusion lncRNA NCK1-AS1 is highly expressed in HCC tissues,which is associated with poor prognosis of patients.It may play a role by regulating the expression of miR-361-5p.
