艾灸百会穴对MCAO大鼠学习记忆能力及海马Bcl-2、Bax的影响 被引量:3

摘要 目的:探究艾灸对MCAO大鼠学习记忆能力及Bcl-2、Bax的影响。方法:30只SD大鼠随机分为假手术组、模型组和艾灸组,每组10只。模型组和艾灸组采用MCAO法复制模型,造模成功后,艾灸组取“百会穴”温和灸,每日1次,每次30 min,共7 d。Morris水迷宫实验观察大鼠学习记忆能力变化,小动物功能核磁共振仪T2加权成像扫描观察大鼠大脑梗死体积,蛋白质免疫印迹法检测Bcl-2、Bax蛋白的表达。结果:定位航行实验,干预第2天模型组和艾灸组逃避潜伏期均长于假手术组,差异均有统计学意义(P<0.05),艾灸组与模型组差异无统计学意义(P>0.05);第3天和第4天艾灸组和假手术组逃避潜伏期短于模型组,差异均有统计学意义(P<0.05),艾灸组与假手术组差异无统计学意义(P>0.05);空间探索实验,艾灸组和假手术组大鼠穿越平台次数多于模型组,差异均具有统计学意义(P<0.05),艾灸组和假手术组差异无统计学意义(P>0.05);艾灸组脑梗死百分比小于模型组[(11.30±0.94)vs(21.74±2.16)%],差异有统计学意义(P<0.05),假手术组Bcl-2/Bax比值最大,其次为艾灸组,最小为模型组;艾灸组与模型组比较差异有统计学意义[(1.71±0.29)vs(0.75±0.15)](P<0.05)。结论:艾灸可明显改善MCAO大鼠的学习记忆能力,缩小脑梗死体积,其机制可能与Bcl-2/Bax增高,抑制神经元凋亡有关。 Objective:To explore the effects of moxibustion on the learning and memory abilities,Bcl-2 and Bax of MCAO rats.Methods:30 SD rats were randomly divided into sham operation group,model group and moxibustion group,with 10 rats in each group.The model group and moxibustion group used MCAO method to replicate the model,and the moxibustion group took mild moxibustion at"Baihui Point"once a day for 7 days.Morris water maze experiment was used to observe the changes in learning and memory ability of rats,small animal functional nuclear magnetic resonance T2-weighted imaging scanning to observe the cerebral infarct volume of rats,and Western blotting to detect the expression of Bcl-2 and Bax protein.Results:In the positioning navigation experiment,on the second day of intervention,the escape latency of the model group and the moxibustion group was longer than that of the sham operation group,and the difference was statistically significant(P<0.05).There was no significant difference between the moxibustion group and the model group(P>0.05);The escape latency of moxibustion group and sham operation group was shorter than model group on the 3rd and 4th day,the difference was statistically significant(P<0.05),the difference between moxibustion group and sham operation group was not statistically significant(P>0.05).In the space exploration experiment,the rats in the moxibustion group and the sham operation group crossed the platform more times than the model group,and the difference was statistically significant(P<0.05).The difference between the moxibustion group and the sham operation group was not statistically significant(P>0.05);the percentage of cerebral infarction in the moxibustion group was less than that in the model group,the difference was statistically significant(P<0.05),the sham operation group had the largest Bcl-2/Bax ratio,followed by the moxibustion group,the smallest was the model group;the difference between the moxibustion group and the model group was statistically significant(P<0.05).Conclusion:Moxibustion can significantly improve the learning and memory ability of MCAO rats and reduce the size of cerebral infarction.The mechanism may be related to the increase of Bcl-2/Bax and inhibition of neuronal apoptosis.
