UFC1在弥漫大B细胞淋巴瘤中的表达研究 被引量:4

摘要 目的研究弥漫大B细胞淋巴瘤(DLBCL)中类泛素结合酶UFC1的表达及意义。方法应用免疫组化法检测UFC1在DLBCL的表达;应用数据库GEPIA和GEO数据集研究UFC1在DLBCL中的表达;应用GEO数据集分析UFC1表达与DLBCL的临床特征的关系;应用GEO数据集及OSdbl分析UFC1表达与DLBCL预后关系;利用Linkedomics分析UFC1共表达基因;应用String-DB分析UFC1相互作用的蛋白网络。结果免疫组化结果显示UFC1在DLBCL中表达明显高于反应性淋巴结炎组织(P<0.001),GEPIA分析显示UFC1在DLBCL表达明显高于正常组织(P<0.01),GSE9327数据集分析也显示UFC1在DLBCL呈高表达(P<0.0001);GSE10846数据集分析显示UFC1高表达与老年(P=0.003)、non-GCB分型(P=0.015)相关,且高表达UFC1的DLBCL患者OS较短(P=0.0326),OSdlc分析显示UFC1高表达的DLBCL患者PFS(P=0.0159)、OS(P=0.0325)均较差。Linkedomics分析显示UFC1与核糖体及氧化磷酸化等多种肿瘤机制相关;String-DB分析显示UFC1与多种肿瘤标志物存在相互作用。结论UFC1在DLBCL中表达升高,UFC1的高表达在老年及non-GCB类型的DLBCL中更明显,UFC1高表达的DLBCL患者可能提示预后不良。UFC1可能成为淋巴瘤潜在治疗靶点。 Objective To study the expression and significance of UFC1 in diffuse large B cell lymphoma(DLBCL).Methods The expression of UFC1 in DLBCL was detected by immunohistochemical method.The expression of UFC 1 in DLBCL was studied by GEPIA and GEO DataSet.The relationship between UFC 1 expression and the clinical characteristics of DLBCL was analyzed by GEO DataSet.The relationship between UFC 1 expression and DLBCL prognosis was analyzed using GEO Dataset and OSdb1.Linkedomics was used to analyze UFC 1 co-expressed genes.String-db was used to analyze the protein network of UFC1 interaction.Results Immunohistochemical results showed that the expression of UFC1 in DLBCL was significantly higher than that of reactive lymphadentis tissue(P=0.000),GEPIA analysis showed that the expression of UFC 1 in DLBCL was significantly higher than that of normal tissue(P<0.01),and GSE9327 Dataset analy sis also showed that UFC1 was highly expressed in DLBCL(P<0.0001).GSE10846 Dataset analys is showed that high expression of UFC1 was associated with old age(P=0.003)and non-GCB typing(P=0.015),and the OS of DLBCL patients with high expression of UFC1 was shorter(P=0.0326),while OSdlc analysis showed that PFS(P=0.0159)and OS(P=0.0325)of DLBCL patients were poor with high expression of UFC1.Linkedomics analysis showed that UFC1 was related to ribosome and oxidative phosphorylation and other tumor mechanisms.String-db analysis shows that UFC 1 interacts with a variety of tumor markers.Conclusion The expression of UFC1 is increased in DLBCL,and the abnormal high expression of UFC1 is more obvious in the elderly and non-GCB type DLBCL.The high expression of UFC1 in patients with DLBCL may indicate poor prognosis.UFC 1 may be a potential therapeutic target for lymphoma.
