摘要
目的:建立高效液相色谱-二极管阵列检测器法(HPLC-DAD)同时测定风热感冒颗粒中苦杏仁苷、(R,S)-告依春、芦丁、连翘酯苷A、3,5-O-二咖啡酰基奎宁酸和牛蒡苷含量的方法。方法:采用液相色谱法,色谱柱为Waters Symmetry ShieldTM RP18,乙腈为流动相A,0.1%磷酸水溶液为流动相B,梯度洗脱;流速为1.0 mL·min-1;柱温为30℃;检测波长为苦杏仁苷和牛蒡苷207 nm,(R,S)-告依春245 nm,芦丁、连翘酯苷A和3,5-O-二咖啡酰基奎宁酸330 nm。结果:苦杏仁苷、(R,S)-告依春、芦丁、连翘酯苷A、3,5-O-二咖啡酰基奎宁酸和牛蒡苷的线性范围分别为23~920(r=0.9994)、0.4~16(r=0.9995)、4.4~176(r=0.9999)、2.8~110(r=0.9998)、5.4~216(r=0.9996)、38~1520μg·mL^(-1)(r=0.9994),加样回收率的平均值(n=6)分别为98.8%、98.7%、98.4%、99.2%、99.8%和98.7%,RSD分别为1.0%、2.5%、1.8%、1.3%、1.3%和0.8%。结论:本法操作简便、结果准确、重复性好,可用于风热感冒颗粒的质量控制。
Objective:To establish an HPLC-DADmethod for determination of amygdalin,(R,S)-epigoitrin,rutin,forsythiaside A,3,5-O-dicaffeoyl quinic acid and arctiin in Fengreganmao granules.Methods:A stable HPLC method was established and the chromatography was accomplished on a Waters Symmetry ShieldTM RP18 column,the mobile phase consisted of acetonitrile(A)-0.1%(w)phosphoric acid(B)as mobile phase in a gradient mode.The flow rate was 1.0 mL·min-1 and the column temperature was set at 30℃.The detection wavelength was set at 207 nm for amygdalin and arctiin,245 nm for(R,S)-epigoitrin,330 nm for rutin,forsythiaside A and 3,5-O-dicaffeoyl quinic acid,respectively.Results:The linear ranges of amygdalin,(R,S)-epigoitrin,rutin,forsythiaside A,3,5-O-dicaffeoyl quinic acid and arctiin were 23-920(r=0.9994),0.4-16(r=0.9995),4.4-176(r=0.9999),2.8-110(r=0.9998),5.4-216(r=0.9996)and 38-1520μg·mL^(-1)(r=0.9994),respectively.The average recovery were 98.8%,98.7%,98.4%,99.2%,99.8%and 98.7%,RSD were 1.0%,2.5%,1.8%,1.3%,1.3%and 0.8%.Conclusion:The method is simple to operate,accurate and reproducible,which can be used for the quality control of Fengreganmao granules.
