摘要
【目的】探究植物生长调节剂对山苍子茎段扦插穗条生长及成活的影响,为完善山苍子快速扦插繁育技术提供一定的参考。【方法】采用单因素随机区组试验,将1年生的山苍子茎段穗条基部分别置于不同质量浓度梯度(1、10、50、100、500和1000 mg/L)的IBA、NAA和ABT溶液中处理1 h,以清水为对照,在沙床上进行扦插试验。每2 d调查穗条发芽率。先筛选生长调节剂相对适宜的质量浓度,再进行L16(43)正交扦插试验,每月调查穗条的成活率。【结果】单因素随机区组扦插试验的结果表明,穗条扦插6~9 d后开始萌芽,且15 d后发芽率达到最高。山苍子茎段穗条发芽率较高的处理:1、10和50 mg/L的IBA处理,发芽率分别为38.00%、51.33%和48.33%,死亡率分别为33.33%、30.00%和44.67%;10、50和100 mg/L的NAA处理,发芽率分别为17.67%、17.67%和20.33%,死亡率分别为69.00%、74.67%和69.33%;1、10和50 mg/L的ABT处理,发芽率分别为40.00%、46.67%和44.67%,死亡率分别为33.33%、52.00%和33.33%。扦插1个月后处理间穗条成活率的差异不显著,平均为97.29%,IBA和ABT对穗条成活的影响程度相当,且均好于NAA。对扦插3个月后穗条发芽率的影响程度由高到低依次为NAA、IBA、ABT,IBA 50 mg/L+NAA 0 mg/L+ABT 50 mg/L处理下穗条的成活率可达50.00%。极差分析结果表明IBA 50 mg/L+NAA 0 mg/L+ABT 25 mg/L为最佳处理组合,50 mg/L的IBA处理下穗条成活率显著高于25和75 mg/L处理,分别为后两者的1.78和2.73倍,但NAA与IBA和ABT对于山苍子穗条扦插成活存在一定的拮抗作用,ABT对山苍子穗条的成活率无显著影响。【结论】使用IBA 50 mg/L+ABT 25~50 mg/L溶液浸泡穗条1 h,可显著促进山苍子茎段穗条成活,扦插1个月后穗条的成活率达97%以上,3个月后成活率可达50.00%以上。
【Objective】The effects of plant growth regulators on the growth and survival of Litsea cubeba stem cuttings were explored,which was an evidence for the rapid cutting propagation technology of L.cubeba.【Method】The single factor randomized block experiment was used and the cutting bottom was soaped into solutions of IBA,NAA and ABT at different mass concentrations(1,10,50,100,500 and 1000 mg/L)for 1 hour,and clear water was CK.The cuttings experiment was carried out on sand bed.The germination rate of cuttings was investigated every 2 days.The suitable mass concentration of growth regulators were screened,followed by L16(43)orthogonal method for the cutting experiment,and the survival rate of cuttings was investigated monthly.【Result】The single factor randomized block experiment showed that cuttings began to sprout after 6 to 9 days,and the sprout rate reached the highest after 15 days.The sprout rate of cuttings were 38.00%,51.33%and 48.33%(death rate were 33.33%,30.00%and 44.67%)under the IBA treatments of 1,10 and 50 mg/L,respectively.The sprout rate were 17.67%,17.67%and 20.33%(death rate were 69.00%,74.67%and 69.33%)under the NAA treatments of 10,50 and 100 mg/L,respectively.The sprout rate were 40.00%,46.67%and 44.67%(death rate were 33.33%,52.00%and 33.33%)under the ABT treatments of 1,10 and 50 mg/L,respectively.The survival rate of each treatment was similar,which average value was 97.29%at the 1st month of cutting,and the effects of IBA and ABT were similar,both of which were better than NAA.While the sprouting influence was NAA>IBA>ABT at the 3rd month of cuttings,and the highest survival rate was 50%under the treatment of IBA 50 mg/L+NAA 0 mg/L+ABT 50 mg/L.However,the treatment of IBA 50 mg/L+NAA 0 mg/L+ABT 25 mg/L was showed the best combination by the range analysis of data.The survival rate of cuttings under 50 mg/L IBA treatment was significantly higher than that of 25 mg/L(1.78 times)and 75 mg/L(2.73 times),while NAA had the antagonistic effect with IBA and ABT on the survival rate of L.cubeba cuttings,ABT had no significant effect on the survival rate of L.cubeba cuttings(P>0.05).【Conclusion】The soaking treatment of IBA(50 mg/L)+ABT(25-50 mg/L)for the stem-cuttings of L.cubeba about 1 hour could be significantly promoted the survival rate of cuttings,which was over 97%in the 1st month and 50.00%more in the 3rd month.
