云南省宣威地区肺癌患者的炎症小体核苷酸结合寡聚化结构域样受体蛋白3基因多态性 被引量:1

Gene polymorphism of inflammasome NOD-like receptor protein 3 among lung cancer patients from Xuanwei,Yunnan Province

摘要 目的探讨云南省宣威地区肺癌患者的炎症小体核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)基因多态性及其临床意义。方法纳入来自云南省宣威地区的80例肺癌患者(观察组)及100例健康体检者(对照组)。收集所有研究对象的基本资料,检测外周血白细胞介素(IL)-1β、IL-18水平,使用TaqMan探针法对NLRP3进行单核苷酸多态性分型。比较两组NLRP3基因位点rs4612666基因型分布和等位基因频率,并采用非条件Logistic回归分析NLRP3基因位点rs4612666与肺癌遗传易感性的相关性。结果观察组血清IL-1β、IL-18表达水平均高于对照组(均P<0.05)。两组NLRP3基因位点rs4612666位点基因型比例以及等位基因频率差异有统计学意义(均P<0.05)。NLRP3基因位点rs4612666 TT基因型为肺癌的易感基因型(OR=1.294,P<0.05),携带NLRP3基因位点rs4612666 T等位基因是肺癌发生的危险因素(OR=1.612,P<0.05)。结论云南省宣威地区肺癌患者的NLRP3基因位点rs4612666 T等位基因频率高于健康人群,炎症小体NLRP3的基因多态性可能与该地区居民肺癌遗传易感性密切相关。 Objective To explore the gene polymorphism of inflammasome NOD-like receptor protein 3(NLRP3)and its clinical significance among lung cancer patients from Xuanwei,Yunnan Province.Methods Eighty lung cancer patients and 100 individuals receiving health examination from Xuanwei,Yunnan Province,were enrolled as observation group and control group respectively.The basic data of all subjects were collected,peripheral blood levels of interleukin(IL)-1βand IL-18 were determined,and single nucleotide polymorphism typing of NLRP3 was performed using TaqMan probe.The genotype distribution and allele frequency of NLRP3 gene locus rs4612666 were compared between the two groups,and the correlation between NLRP3 gene locus rs4612666 and genetic susceptibility to lung cancer was analyzed using the unconditional Logistic regression model.Results The expression levels of serum IL-1βand IL-18 in the observation group were higher than those in the control group(all P<0.05).There were statistically significant differences between the two groups in genotype proportion or allele frequency of NLRP3 gene locus rs4612666(all P<0.05).TT genotype of NLRP3 gene locus rs4612666 was the susceptible genotype of lung cancer(OR=1.294,P<0.05),and carrying T allele of NLRP3 gene locus rs4612666 was the risk factor of developing lung cancer(OR=1.612,P<0.05).Conclusion Lung cancer patients from Xuanwei,Yunnan Province,possess a higher frequency of T allele of NLRP3 gene locus rs4612666 than healthy population,and gene polymorphism of inflammasome NLRP3 may be closely related to genetic susceptibility to lung cancer of the residents in this area.
