辽宁省野生羊肚菌资源调查与遗传多样性分析 被引量:6

摘要 为了调查辽宁省的羊肚菌资源,了解其产地环境及种属的关系,探寻羊肚菌在辽宁省发生的适合条件,对辽宁省7市16个羊肚菌发生地开展了资源调查,共采集到羊肚菌样本53份。采用ITS和多基因系谱一致性系统发育种识别法(GCPSR),对采集到的羊肚菌样本进行鉴定和遗传多样性分析。结果表明,采集的羊肚菌样本被鉴定为5种,即Morchella sp.(Mes-6)、Morchella sp.(Mes-8)、Morchella sp.(Mes-9)、Morchella sp.(Mes-20)、Morchella sp.(Mes-21),其中,Mes-20和Mes-21为辽宁省新记录种。辽宁省的野生羊肚菌资源主要集中在朝阳市,以Mes-6为主。采用11条ISSR引物,共检测出多态位点51个。在居群水平上,Nei’s基因多样性为0.1201±0.0466,居群间遗传变异占总变异的60.79%,居群内占39.21%,遗传变异主要存在于居群间。 In order to investigate Morchella spp.resources to understand the environment of origin and species relations,find suitable conditions for Morchella spp.in Liaoning province.A habit survey of Morchella spp.was conducted in 7 cities,Liaoning,China.53 specimens were collected in 16 locations.By ITS and genealogical concordance phylogenetic species recognition(GCPSR),the samples were identified as Morchella sp.(Mes-6),Morchella sp.(Mes-8),Morchella sp.(Mes-9),Morchella sp.(Mes-20),Morchella sp.(Mes-21).Morchella sp.(Mes-20)and Morchella sp.(Mes-21)were first reported in Liaoning province.Wild Morchella spp.resources were mainly Mes-6 in Liaoning.Eleven ISSR primers were screened for the amplification of Morchella spp.,51 polymorpha bands were resulted.At the population level,Nei’s gene diversity was 0.1201±0.0466.The interpopulational variation accounted for 60.79%,and intrapopulational variation 39.21%.Moreover,the gene differentiation was existed in Morchella spp.interpopulations.
