成纤维细胞生长因子受体2剪切突变体Ⅲc在膀胱癌中的表达以及与膀胱癌253J细胞多柔比星耐药的关系 被引量:1

Expression of FGFR2 Ⅲc in bladder cancer and its relationship with doxorubicin resistance in 253J bladder cancer cells

摘要 目的探讨成纤维细胞生长因子受体2的剪切突变体Ⅲc(FGFR2Ⅲc)与膀胱癌发生发展及膀胱癌253J细胞多柔比星耐药的关系。方法选取2016年1月至2018年1月郑州市第一人民医院保存的膀胱癌组织标本143例,同时选取正常膀胱组织70例作为对照,采用实时荧光定量聚合酶链式反应(qRT-PCR)检测组织FGFR2Ⅲc表达;选取膀胱癌253J细胞,同时建立耐多柔比星253J细胞,采用MTT法检测最大半数抑制浓度,细胞划痕试验检测细胞迁移。结果膀胱癌组FGFR2Ⅲc mRNA相对表达量为(1.782±0.342),明显高于正常膀胱组(P<0.05);病理分级高级别、病理分期T2~T4期FGFR2Ⅲc mRNA相对表达量分别为(1.945±0.330)和(1.869±0.322),明显高于低级别和Ta~T1期(P<0.05);不同病理分级、病理分期病人性别、年龄比较差异无统计学意义(P>0.05);耐多柔比星253J细胞FGFR2Ⅲc mRNA相对表达量和最大半数抑制浓度分别为(2.001±0.291)和(13.02±1.10)g/L,明显高于253J细胞(P<0.05);耐多柔比星253J细胞迁移率为(23.02±3.12)%,明显低于253J细胞(P<0.05);耐多柔比星253J细胞凋亡率为(23.22±5.50)%,明显低于253J细胞(P<0.05)。结论FGFR2Ⅲc mRNA在膀胱癌组织中表达上调,与病理分级和病理分期有关,同时与253J细胞耐多柔比星、迁移有一定关系。 Objective To investigate the relationship between the shear mutant of fibroblast growth factor receptor 2(FGFR2Ⅲc)and the occurrence and development of bladder cancer and the drug resistance of bladder cancer 253J cells to doxorubicin.Methods 143 specimens of bladder cancer were collected from January 2016 to January 2018,and 70 normal bladder tissues were selected as controls,the expression of FGFRR2Ⅲc mRNA was detected by qRT-PCR;Bladder cancer 253J cells were selected and doxorubicinresistant 253J cells were established,MTT assay was used to detect the maximum inhibitory concentration,and cell scratch test was used to detect cell migration.Results The relative expression of FGFR2Ⅲc in bladder cancer group was(1.782±0.342),which was significantly higher than that in normal bladder group(p<0.05);The relative expression levels of FGFR2Ⅲc in high grade and stage T2-T4 were(1.945±0.330)and(1.869±0.322),which were significantly higher than those in low grade and stage Ta-T1(P<0.05);There were no statistically significant differences in gender and age among patients with different pathological grades and stages(P>0.05);The relative expression of FGFR2Ⅲc and the maximum half inhibitory concentration of FGFR2Ⅲc in doxorubicin-resistant253J cells were(2.001±0.291)and(13.02±1.10)g/L,which were significantly higher than those in 253J cells(P<0.05);The migration rate of doxorubicin-resistant-253J cells was(23.02±3.12)%,which was significantly lower than that of 253J cells(P<0.05);The apoptosis rate of doxorubicin 253J cells was(23.22±5.50)%,which was significantly lower than that of 253J cells(P<0.05).Conclusion The expression of FGFR2Ⅲc is up-regulated in bladder cancer tissues,which are related to pathological grading and stage,and also to doxorubicin resistance and migration of 253J cells.
