摘要
目的以青砖茶为原料,探究提取温度对青砖茶多糖组分含量和生物活性的影响。方法采取20℃、40℃、60℃、80℃及100℃5个提取温度,分别水提醇沉淀制备青砖茶粗多糖,经聚酰胺层析柱除去粗多糖里的杂质,得到20℃提取青砖茶多糖(TPS-20)、40℃提取青砖茶多糖(TPS-40)、60℃提取青砖茶多糖(TPS-60)、80℃提取青砖茶多糖(TPS-80)及100℃提取青砖茶多糖(TPS-100)。再通过苯酚浓硫酸法、高效液相色谱-质谱联用(HPLC-MS)法、1,1-二苯基-2-苦肼基(DPPH)自由基清除活性实验、羟自由基清除活性试验、抑制α-葡萄糖苷酶活性实验研究这五个多糖组分的得率、糖含量、单糖组成、抗氧化活性及抑制α-葡萄糖苷酶活性。结果TPS-20、TPS-40、TPS-60、TPS-80及TPS-100的得率分别为(2.02±0.02)%、(2.94±0.05)%、(3.18±0.07)%、(3.03±0.09)%及(2.11±0.06)%;糖含量分别为(36.87±2.39)%、(44.71±2.91)%、(52.76±4.24)%、(52.33±4.01)%及(35.93±1.73)%,均含有核糖、木糖、阿拉伯糖、鼠李糖、岩藻糖、甘露糖、葡萄糖、半乳糖、半乳糖醛酸;不同提取温度多糖均具有1,1-二苯基-2-苦肼基(DPPH)自由基清除活性、羟自由基清除活性、抑制α-葡萄糖苷酶活性,其中40到80℃之间提取得到的青砖茶多糖的DPPH自由基清除活性更优。结论在一定温度范围内,提取温度对青砖茶多糖单糖的种类及抑制α-葡萄糖苷酶活性没有显著影响,对青砖茶多糖得率、糖含量、抗氧化活性有一定的影响。TPS-60的得率及糖含量均较高,且具有抗氧化活性及抑制α-葡萄糖苷酶活性,即60℃提取得到的青砖茶多糖具有潜在的开发应用价值,有望作为天然抗氧化剂及治疗糖尿病的药物加以研发利用。
Objective The study is to explore the effects of different extraction temperature on component content and bioactivities of the polysaccharides from Qingzhuan Dark Tea.Methods We gained five types of crude tea polysaccharides(CTPS)by precipitating the water extract of Qingzhuan Dark Tea with different extraction temperature,20℃,40℃,60℃,80℃and 100℃.Furthermore,we purified those five CTPS named TPS-20,TPS-40,TPS-60,TPS-80 and TPS-100,by polyamide column chromatography.We investigated the extraction rates,contents of carbohydrate,monosaccharides composition,antioxidant activities and inhibitory activities toα-glucosidase of those TPS by using phenol-sulfuric acid method,HPLC-MS(High Performance Liquid Chromatography-Mass Spectrometry)method,assay of DPPH radical scavenging activity,assay of hydroxy radical scavenging activity and assay of inhibitory activity toα-glucosidase.Results The extraction rates of TPS-20,TPS-40,TPS-60,TPS-80 and TPS-100 were(2.02±0.02)%,(2.94±0.05)%,(3.18±0.07)%,(3.03±0.09)%and(2.11±0.06)%.The carbohydrate contents of those TPS were(36.87±2.39)%,(44.71±2.91)%,(52.76±4.24)%,(52.33±4.01)%and(35.93±1.73)%.There were mannose,ribose,arabinose,rhamnose,xylose,galacturonic acid,fucose,galactose and glucose in all types of polysaccharide.All TPS with different extraction temperatures had DPPH(1,1-diphenyl-2-picrylhydrazyl)radical scavenging activity,hydroxy radical scavenging activity and the inhibitory activity toα-glucosidase.The DPPH radical scavenging activity of TPS-40,TPS-60 and TPS-80 were better than that of TPS-20 and TPS-100.Conclusion The extraction temperature had no effect on monosaccharides composition and inhibitory activities toα-glucosidase of the polysaccharides from Qingzhuan Dark Tea in a certain temperature range,but had effects on extraction rates,contents of carbohydrate and antioxidant activities.The extraction rate and carbohydrate content of TPS-60 were higher than others,and TPS-60 has antioxidant activities and inhibitory activities toα-glucosidase.Which means that TPS-60 possesses potential value for drug development and utilization.
