摘要
目的:基于糖尿病并发症探讨紫荆花总黄酮对2型糖尿病模型大鼠的药效作用研究。方法:采用喂养高脂高糖饲料4周加一次性腹腔注射链脲菌素(65 mg·kg^(-1))建立2型糖尿病大鼠模型,一周后测定空腹血糖值,选取血糖值在16.7~21.0 mmol·L^(-1)的大鼠,随机分为模型组、盐酸二甲双胍组(0.35 g·kg^(-1))、紫荆花总黄酮(高、低剂量)(200 g·kg^(-1)、100 g·kg^(-1))组,另设空白组,各组大鼠连续给予相应药物灌胃30 d。每2周测定空腹血糖值;腹主动脉取血,采用酶联免疫法测定血清糖化血清蛋白(glycated serum proteins,GSP)、胰岛素(insulin,INS)、胰岛素自身抗体(insulin autoantibody,IAA)、总胆固醇(total cholesterol,TC)、三酯甘油(triglyceride,TG)、高密度脂蛋白(high density lipoprotein,HDL-C)、低密度脂蛋白(low density lipoprotein,LDL-C)、总抗氧化能力(total antioxidant capacity,T-AOC)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)水平;全血检测血液流变学相关指标。结果:与模型组相比,紫荆花总黄酮组均可不同程度降低模型大鼠空腹血糖值;降低血清GSP、IAA、TC、TG、LDL-C、MDA水平,升高INS、HDL-C、SOD、T-AOC水平;对血液流变学相关指标也有很好的改善作用。结论:紫荆花总黄酮可有效调节2型糖尿病大鼠血清血脂水平紊乱,增强机体抗氧化能力,有效降低空腹血糖值,达到良好降糖效果。
Objective:To investigate the pharmacological mechanism of total flavonoids of bauhinia chinensis on type 2 diabetic rats based on diabetic complications.Methods:The animal model of type 2 diabetic was established by feeding high fat and high sugar diet for 4 weeks and intraperitoneal injection of STZ 65 mg·kg^(-1).The rats with blood glucose value of 16.7-21.0 mmol·L^(-1) were randomly divided into model group,metformin hydrochloride group,metformin hydrochloride group(0.35 g·kg^(-1)),total flavonoids of Bauhinia chinensis(high and low dose)(100 g·kg^(-1),200 g·kg^(-1))groups,and blank group.The rats in each group were given corresponding drugs by gavage for 30 days.Fasting blood glucose was measured every 2 weeks;blood samples were collected from abdominal aorta and levels of serum glycated serum proteins(GSP),insulin(INS),insulin autoantibody(IAA),total cholesterol(TC),triglyceride(TG),high density lipoprotein(HDL-C),low density lipoprotein(LDL-C),total antioxidant capacity(T-AOC),superoxide dismutase(SOD)and malondialdehyde(MDA)were determined by enzyme-linked immunosorbent assay;hemorheology related indexes were detected in whole blood.Results:Compared with the model group,the total flavonoids of Bauhinia chinensis group can reduce the fasting blood glucose,reduce the serum GSP,IAA,TC,TG,LDL-C,MDA levels,increase INS,HDL-C,SOD,T-AOC levels;and improve the hemorheology related indexes.It can also improve the hemorheology related indexes.Conclusion:The total flavonoids of bauhinia chinensis can effectively regulate the disorder of serum lipid level in type 2 diabetic rats,enhance the body's antioxidant capacity,effectively reduce fasting blood glucose,and achieve good hypoglycemic effect.
