绞股蓝多糖对断乳雌性小鼠卵巢发育的影响 被引量:4

Effects of Gynostemma pentaphyllum polysaccharide on ovarian development in weaned female mice

摘要 为了探究不同剂量的绞股蓝多糖(Gynostemma pentaphyllum polysaccharides,GPP)对断乳雌性小鼠卵巢发育的影响,本研究采用酶联免疫吸附和石蜡切片等研究方法,分别对用药第7和第14天小鼠的体重变化,卵巢和子宫指数,阴道开放情况,血清中促性腺激素释放激素(gonadotropin releasinghormone,GnRH)、促卵泡素(follicle stimulating hormone,FSH)、促黄体素(luteinizing hormone,LH)、雌二醇(estradiol,E2)和孕酮(progesterone,PG)的含量以及卵巢中各级卵泡数量、最大直径及卵巢皮质厚度进行测定分析。结果显示,不同剂量的GPP均可促进小鼠体重增加,使小鼠阴道开放时间提前,卵巢和子宫指数增加,促进小鼠GnRH、FSH、LH和E2的分泌,抑制PG的分泌,促进卵巢各级卵泡的生长发育,使小鼠卵巢皮质厚度显著增加。其中用药第14天时,GPP-L组小鼠体重显著高于空白对照组(P<0.05);GPP-M组小鼠阴道开放率显著高于空白对照组与GPP-H组(P<0.05),GPP-M组小鼠阴道开放时间较GPP-L和GPP-H组显著提前(P<0.05);GPP-L和GPP-M组小鼠血清中GnRH、FSH、LH和E2的含量均显著高于空白对照组(P<0.05);GPP-L和GPP-M组小鼠卵巢成熟卵泡数和黄体数均显著高于空白对照组(P<0.05);GPP-M组小鼠卵巢原始卵泡数显著低于GPP-H组(P<0.05);GPP-M组小鼠卵巢原始卵泡、初级卵泡、次级卵泡和黄体最大直径显著大于空白对照组和GPP-H组(P<0.05);GPP-L和GPP-M组小鼠卵巢皮质厚度显著高于空白对照组和GPP-H组(P<0.05)。上述结果表明,用药第14天时,GPP-L(50 mg/kg)组效果明显,GPP-M(100 mg/kg)组效果最佳,该研究结果将为绞股蓝多糖进一步研究与开发利用提供新的方向与理论依据。 In order to explore the effects of different doses of Gynostemma pentaphyllum polysaccharides(GPP),enzyme linked immunosorbent assay(ELISA),paraffin section and other research methods were used to determine the weight change,ovarian and uterus index,vaginal opening,GnR H,FSH,LH,E2 and PG contents in serum,as well as the number of follicles at all levels,the maximum diameter and the thickness of ovarian cortex on the 7 th and 14 th days of administration.The results showed that different doses of GPP could promote the weight gain in mice,advance the opening time of vagina,increase the ovarian index and uterine index,promote the secretion of GnR H,FSH,LH and E2,inhibit the secretion of PG,promote the growth and development of ovarian follicles at all levels,and significantly increase the thickness of ovarian cortex. On the 14 th day of administration,the weight of mice in GPP-L group was significantly higher than that in blank control group(P<0.05);vaginal opening rate of GPP-M group was significantly higher than that of blank control group and GPP-H group(P<0.05);the vaginal opening time of GPP-M group was significantly earlier than that of GPP-L group and GPP-H group(P<0.05);GnR H,FSH,LH and E2 in serum of GPP-L group and GPP-M group were significantly higher than those in blank control group(P<0.05);the number of mature follicles and corpus luteum in GPP-L group and GPP-M group were significantly higher than those in blank control group(P<0.05);the number of primordial follicles in GPP-M group was significantly lower than that in GPP-H group(P<0.05);the maximum diameters of primordial follicle,primary follicle,secondary follicle and corpus luteum in group GPP-M were significantly larger than those in blank control group and GPP-H group(P<0.05);the thickness of ovarian cortex in GPP-L group and GPP-M group was significantly higher than that in blank control group and GPP-H group(P<0.05). According to the results,the effect of GPP-L(50 mg/kg) was obvious,and that of GPP-M(100 mg/kg) was the best on the 14 th day of administration.It will provide a new direction and theoretical basis for further research and development and utilization of Gynostemma pentaphyllum polysaccharide.
