MEI1在宫颈癌中高表达并促进宫颈癌细胞的增殖与迁移侵袭 被引量:2

High Expression of MEI1 in Cervical Cancer Promotes Proliferation and Migration of Cervical Cancer Cells

摘要 目的探究减数分裂抑制物1(meiosis inhibitor 1,MEI1)对宫颈癌细胞增殖以及迁移、侵袭能力的影响。方法在GEPIA数据库中分析MEI1在宫颈癌组织和正常组织中的表达量,以及MEI1表达对宫颈癌患者总体生存期的影响;以荧光实时定量聚合酶链反应(quantitative real-time polymerase chain reaction,qRT-PCR)、免疫组化以及Western blot方法检测MEI1在宫颈癌组织及正常组织中的表达;构建过表达MEI1的ME-180-MEI1细胞和敲减MEI1的HCC-94-shMEI1细胞,通过CCK-8以及平板克隆形成实验、划痕实验和Transwell实验分别检测MEI1对宫颈癌细胞增殖和迁移侵袭的影响;qRT-PCR以及Western blot检测MEI1对Cyclin D1、增殖细胞核抗原(PCNA)、金属基质蛋白(MMP)、钙粘蛋白(cadhexin)以及波形蛋白(Vimentin)表达的影响。结果人宫颈癌组织中MEI1 mRNA的表达水平明显高于宫颈正常组织,MEI1高表达的患者其总体生存较差(P<0.01)。CCK-8以及平板克隆形成实验证明MEI1过表达可明显增加宫颈癌细胞的增殖(P<0.01),而敲减MEI1则降低宫颈癌细胞的增殖(P<0.01);划痕实验及Transwell实验证明MEI1过表达显著增强宫颈癌细胞的迁移侵袭能力(P<0.01),而敲减MEI1则明显降低宫颈癌细胞的迁移侵袭能力(P<0.01);qRT-PCR以及Western blot实验证明过表达MEI1可以促进Cyclin D1、PCNA、MMP-2、MMP-9、MMP-14、E-cadherin、N-cadherin、Vimentin的表达,而敲减MEI1则可明显抑制宫颈癌细胞中细胞周期蛋白的表达,并影响其上皮间质转化(EMT)相关基因的表达。结论MEI1在宫颈癌组织中表达显著高于正常组织,其可能通过影响细胞周期蛋白以及EMT相关蛋白的表达,进而促进宫颈癌细胞的增殖以及迁移、侵袭能力。 Objective To explore the effect of meiosis inhibitor 1(MEI1)on proliferation and migration of cervical cancer cells.Methods The expression of MEI1 in cervical cancer tissues and normal tissues was determined in the GEPIA analysis database,and the effect of MEI1 expression on the overall survival of patients with cervical cancer was analyzed.The expression of MEI1 in cervical cancer and normal tissues was detected by quantitative real-time polymerase chain reaction(qRT-PCR),immunohistochemistry and Western blotting.Overexpressing of MEI1 in ME-180-MEI1 cells and knocking down of MEI1 in HCC-94-shMEI1 cells were constructed to detect the effect of MEI1 on proliferation,migration and invasion of cervical cancer cells by CCK-8 as well as plate colony formation assay,scratch assay and transwell assay,respectively;the effect of MEI1 on the expression of Cyclin D1,PCNA,matrix protein,cadherin and vimentin was detected by qRT-PCR and Western blotting.Results The mRNA level of MEI1 in human cervical cancer tissues was significantly higher than that in normal cervical tissues.Patients with high MEI1 expression had poorer overall survival(P<0.01).CCK-8 and plate clone formation assay proved that overexpression of MEI1 significantly increased the proliferation of cervical cancer cells(P<0.01),while knocking down of MEI1 reduced the proliferation of cervical cancer cells(P<0.01).The scratch assay and transwell assay showed that MEI1 overexpression significantly enhanced the migration and invasion ability of cervical cancer cells(P<0.01),while knocking down of MEI1 significantly reduced the migration and invasion of cervical cancer cells(P<0.01).qRT-PCR and Western blotting proved that overexpression of MEI1 promoted the expression of Cyclin D1,PCNA,MMP-2,MMP-9,MMP-14,E-cadherin,N-cadherin,and Vimentin,while knocking down of MEI1 significantly inhibited the expression of Cyclin in cervical cancer cells and affected their epithelial mesenchyme transformation(EMT)related gene expression.Conclusion The expression of MEI1 in cervical cancer tissues was significantly higher than that in normal tissues.MEI1 might promote the proliferation,migration and invasion of cervical cancer cells by affecting the expression of Cyclin and EMT-related proteins.
