摘要
目的研究hsa-miR-223-3p和hsa-miR-33b-3p在川崎病患儿外周血中表达的意义。方法选取2018年6月-2019年5月永康市中医院和浙江大学医学院附属儿童医院收治的川崎病患儿8例为观察组,同期在该院体检的健康儿童8例为对照组。提取两组研究对象血浆中总RNA,采用miRNA芯片杂交技术检测两组研究对象差异表达的miRNAs,采用RT-qPCR技术验证芯片结果。结果 miRNA芯片中hsa-miR-223-3p显著上调表达,hsa-miR-33b-3p显著下调表达,差异均有统计学意义(均P<0.05)。hsa-miR-223-3p在川崎病血管炎症中具有保护作用,可以减轻血管炎症。结论川崎病患儿外周血中hsa-miR-223-3p和hsa-miR-33b-3p表达异常,其参与了川崎病的病理生理过程。
Objective To investigate the expression significance of hsa-miR-223-3p and hsa-miR-33b-3p in the peripheral blood of children with Kawasaki disease.Methods Eight children with Kawasaki disease who were admitted to Yongkang Traditional Chinese Medicine Hospital and the Children’s Hospital Zhejiang University School of Medicine from June,2018 to May,2019 were selected as the observation group,and eight healthy children who underwent physical examination in the hospital during the same period were selected as the control group.Total RNA was extracted from the plasma of the subjects in the two groups,miRNA microarray hybridization was used to detect the differentially expressed miRNAs of the subjects in the two groups,and RT-qPCR was used to verify the microarray results.Results The expression of hsa-miR-223-3p in miRNA microarray was significantly up-regulated,and the expression of hsa-miR-33b-3p was significantly down-regulated,the differences were statistically significant(all P<0.05).Hsa-miR-223-3p has a protective effect on vascular inflammation in Kawasaki disease,and can reduce vascular inflammation.Conclusion The abnormal expression of hsa-miR-223-3p and hsamiR-33b-3p in the peripheral blood of children with Kawasaki disease is involved in the pathophysiological process of Kawasaki disease.
