摘要
建立测定虎眼万年青叶、茎中木犀草素含量的方法,并研究其体外抗氧化活性。采用反相高效液相色谱法对样品甲醇提取物进行分析。色谱柱为Agilent Eclipse XDB-C_(18)(4.6 mm×250 mm,5μm);流动相为甲醇-0.1%磷酸水溶液(55∶45,V∶V);流速为1.0 mL/min;柱温为30℃;检测波长为275 nm。结果表明,木犀草素浓度在0.3236~16.18μg/mL范围内呈良好的线性关系,平均加样回收率为98.31%,RSD为1.98%。测得虎眼万年青叶和茎中木犀草素的平均含量分别为0.0149 mg/g和0.0198 mg/g。虎眼万年青提取物能显著抑制DPPH自由基、ABTS自由基的产生,具有良好的抗氧化活性。
This paper established a method for the determination of luteolin in leaves and stems of Ornithogalum caudatum,and studied its antioxidant activity in vitro.The methanol extract was analyzed by RP-HPLC.The determination was performed on Agilent Eclipse XDB-C_(18)(4.6 mm×250 mm,5μm)column with mobile phase consisted of methanol-0.1%phosphoric acid solution(55∶45,V∶V)at the flow rate of 1.0 mL/min.The column temperature was 30℃and the detection wavelength was 275 nm.The results showed that the concentration of luteolin showed a good linear relationship in the range of 0.3236~16.18μg/mL,the average recovery was 98.31%,RSD was 1.98%.The average contents of luteolin in leaves and stems of Ornithogalum caudatum were 0.0149 mg/g and 0.0198 mg/g.The extract of Ornithogalum caudatum could significantly inhibit the production of DPPH and ABTS free radicals,it showed good antioxidant activity.
