微小核糖核酸-222过表达的心肌干细胞移植对急性心肌梗死大鼠心功能的影响 被引量:1

摘要 目的:探讨miR-222过表达的心肌干细胞(CSC)移植对急性心肌梗死(AMI)大鼠心功能的影响.方法:SD大鼠40只,随机分为假手术组、模型组、阴性对照组和模拟物组,每组10只,构建大鼠AMI模型,分别在阴性对照组和模拟物组梗死区域注射对照序列或miR-122-CSC细胞悬液.建模后4周,检查各组大鼠心脏功能,HE染色和Masson染色观察各组大鼠心肌组织形态学变化,实时荧光定量PCR检测各组大鼠心肌梗死组织中miR-222 mRNA表达,TUNEL法检测各组大鼠心肌梗死组织中细胞凋亡,Western blot法检测各组大鼠心肌梗死组织中Caspase-3和p-H3蛋白表达.结果:与假手术组相比,模型组、阴性对照组和模拟物组大鼠心脏LVEDD和LVESD升高,而LVEF降低,心肌梗死组织中miR-222 mRNA的相对表达量升高,细胞凋亡指数升高,Caspase-3和p-H3蛋白相对表达量升高,差异有统计学意义(P<0.05);与模型组和阴性对照组相比,模拟物组大鼠心脏LVEDD和LVESD降低,而LVEF升高,心肌梗死组织中miR-222 mRNA的相对表达量升高,细胞凋亡指数降低,Caspase-3蛋白相对表达量降低,而p-H3蛋白相对表达量升高,差异有统计学意义(P<0.05).结论:miR-222过表达的CSC移植可改善AMI大鼠心脏功能,其机制可能与抑制心肌细胞凋亡,促进细胞增殖有关. Objective:To investigate the effect of miR-222 over-expressed cardiac stem cells(CSC)transplantation on cardiac function after acute myocardial infarction(AMI).Methods:CSCs were isolated and cultured from heart of newborn SD rat,and were transfected with miR-122 mimics(miR-122-CSC group)and negative control sequences(control sequence-CSC group),respectively.A total of 60 SD rats were randomly di-vided into sham group,model group,negative control group and mimics group,each group had 10.Rat model of AMI was constructed.1×10^(6)cells suspensions from miR-122-CSC group or control sequence-CSC group were injected into the infarct areas of the negative control group and the mimics group,respectively.4 weeks after modeling,the heart function of rats in each group was tested.HE staining and Masson staining were used to ob-serve the histomorphological changes in myocardial tissues in each group.Real-time fluorescence quantitative PCR was used to detect the expressions of miR-222 in myocardial infarction tissues of rats in each group.TUNEL assay was used to detect the cell apoptosis in myocardial infarction tissues of rats in each group.West-ern blot was used to detect the expressions of Caspase-3 and p-H3 proteins in myocardial infarction tissues of rats in each group.Results:Compared with the sham group,in the model group,negative control group and mimics group,the LVEDD and LVESD were increased,while the LVEF was decreased,the relative expression levels of miR-222 in the myocardial infarction tissues were increased,the apoptosis index was increased,and the relative expression levels of Caspase-3 and p-H3 proteins were increased,the differences were statistically significant(P<0.05).Compared with the model group and the negative control group,in the mimics group,the LVEDD and LVESD were decreased,while the LVEF was increased,the relative expression of miR-222in the myocardial infarction tissues was increased,the apoptosis index was decreased,and the relative expression level of Caspase-3 protein was decreased,while the relative expression level of p-H3 protein was increased,the differences were statistically significant(P<0.05).Conclusions:MiR-222 over-expressed cardiac stem cells transplantation could improve cardiac function in rat with AMI.The mechanism might be related to inhibition of myocardial cell apoptosis and promotion of cell proliferation.
